Objective:To determine the effects of ursolic acid(UA),zedoary oil and zedoary oil jointγ-ray irradiation on the proliferation and apoptosis in human nasopharyngeal carcinoma(NPC)cell lines CNE-2 and explore its underlying mechanisms.Analysis of whether the zedoary oil enhancement of the role of radiotherapy.Methods:UA or zedoary oil was added into the cultured CNE-2 cells and incubated for 24,48,72,96and 120h.MTT assay and Flow cytometry were employed to evaluate the proliferation,apoptosis and cell cycle of CNE-2 cells. Apoptotic bodies were observed by electronic and Fluorescence microscopeResults:Both UA and zedoary oil can inhabit the proliferation and induce the apoptosis of the CNE-2 cell when used alone,which is similar to the effect of 5-FU(P>0.05)(at the set Concentration of drugs in our experiment).UA exerts the inhibiting effect in a dose -and time -dependent manner,the lowest effect concentration is 10μmol/ml,the optimal time is 72h;while zedoary oil inhabit the cell proliferation in a concentration -dependent manner,the lowest effect concentration and the optimal time is 10μg/ml;combining zedoary oil(5μg/ml,100μg/ml and 300μg/ml) for 48h,the apoptosis rate of CNE-2 cell was 0.50%,2.15%,10.2%respectively,while combined withγ-ray(100cGY),the apoptosis rate increased to 8.6%,14%,26%respectively,the average mortality rate of cell was below 5%,the synergy effect was obvious(P<0.01) when combined for 96h,the experiment group cells died mainly,but few apoptosis. High concentration zedoary oil(500μg/ml) had an obvious cytotoxic effect on cells,and finally cause cell to die.Flow cytometry indicated that CNE-2 was blocked at the G1 phase of the cell cycle when UA and zedoary oil was at a high concentration.Apoptotic bodies were observed in both of the drug treated CNE-2 cells by electron microscope and Fluorescence microscopeConclusion:Both of UA and zedoary oil inhabit the proliferation of CNE-2 and induce cell apoptosis of the CNE-2 cell;when combined withγ-ray,zedoary oil enhance the sensitivity of radio therapy significantly.Its mechanism of induction of apoptosis may be related to the impact of cell cycle-related.
