The Effects Of Venom Nerve Growth Factor On GAP-43 Of Nerve Cell Following Cerebal Ischemia Injured In Rat

Objective:Applying focal ischemia reperfusion model of rat,to observe the change of expression of GAP-43(growth associated protein-43) and neurological function after 7 days.To investigating the cerebral ischemial reperfusion protection mechanism of vNGF(viper venom nerve growth factor) and to offer the theory of curing ischemic cerebrovascular disease.Methods:45 adult male Wistar rats(weight 220-280g) were divided randomly into 3 groups:sham group(S)(n=9),balanced salt solution group(BSS)(n=9) and vNGF group(n=27)。Each group were observed 7 days. vNGF group were divided into 25u,50u and 100u subgroup respectively,and each subgroup had 9 rats.The following indexes in 3 groups were observed respectively:neurologic deficits,tissue pathological changes by Hematoxylin-Eosin(H.E) staining,and the expression of GAP-43 (immunohistochemistry method).Results:(1) Neurological function:The scores of neurological function were 0 in S group.The neurological deficits score is lower at the same time in vNGF group than that in BSS group(P＜0.05).(2) H.E stain showed that the pictures under substantial microscope showed that S group had no evident pathological changes.Infarction center of vNGF group decreased distinctively compared with BSS group,structure integrated neural cells around infarction areas increased cleared.(3) Immunohistochemistry:excepted S group,both BSS group and vNGF group had GAP-43 expression.The expression of GAP-43 in vNGF group were increase in 25u,and to maxximum in 100u.The expression of GAP-43 in BSS group were significantly lower than vNGF group (P＜0.05)Conclusion:After cerebral ischemia injured,vNGF are effective in enhancing and prolonging the expression of GAP-43,increasing the survival rats of nerve cells,and has the protection effect on nerve cells.They had dose-effect relationship between nerve cells protection effect and GAP-43.
