Objective: To investigate the relationship between HLA-DRB1*07, 15 gene and the effect of immune response to hepatitis B vaccine among Zhuang and Han nationality in Guangxi, and the impact of nationality`s distinction to the relationship between HLA-DRB1*07, 15 gene and the effect of immune response to hepatitis B vaccine.Methods: 684 healthy college students of Zhuang or Han nationality from Guangxi province who had received standard courses of vaccination with recom -binant heaptitis B virus vaccine ,were tested HBV makers (HBsAg, anti-HBs, HBeAg, anti-HBe, anti-HBc) and liver function in half a year after their vaccination. 83 non- or hypo-responders (serum Anti-HBs<10mIu/ml) and 166 medium or hyper-responders (serum Anti-HBs≥10 mIu/ml) with normal liver function and negative HBV makers of those students were selected randomly and involved in this study. Detection of HLA-DR B1*07, HLA-DRB1*15 were performed by polymerase chain reaction using sequence specific primer (PCR-SSP). Analyze the relationship between HLA-DRB1*07, 15 gene and immune response to hepatitis B vaccine in either nationality, and the relationship between nationality, HLA-DRB1*07, 15 gene and the effect of immune response were analyzed. Results: In either Zhuang or Han nationality, the frequency of medium or hyper-responders in the carriers of HLA-DRB1*07 gene was significantly lower than that of non HLA-DRB1*07 carriers, the odd ratio (OR) was 0.193(P=0.019),and 0.167(P=0.000) respectively; The frequency of medium or hyper-res- ponders in the carriers of HLA-DRB1*15 gene was higher than that of non-HLA -DRB1*15 gene carriers, the odd ration(OR) was 3.476(P=0.024)and 2.891 (P=0.006). No significant difference was shown between the frequencies of HLA-DRB1*07, 15 in Zhuang and Han nationality (P=0.899/0.788). There was also no significant difference between the Zhuang and Han nationality in the frequencies of immune-responders (P=0.55/ 0.413).Conclusions: HLA-DRB1*07 gene may be a factor that associated with the non or hepo-response to hepatitis B vaccine in Zhuang or Han nationality from Guangxi province and HLA-DRB1*15 gene is associated with the medium or hyper-response to hepatitis B vaccine. The distinction of the nationality is not the factor which impacts on the relationship between HLA-DR B1*07, 15 gene and immune response to hepatitis B vaccine .
