Objective: To investigate the effect of Alprostadil postconditioning on myocardial ischemia-referfusion injury in rats.Methods: The rat models of myocardial ischemia-reperfusion was established with occlusion of the left anterior descending coronary artery(LAD) for 30 min and reperfusion for 60min. 32 rats were divided into 4 groups, group A of them was taken as sham operation group, group B control group (ischemia 30 min, reperfusion 60 min), group C(Alprostadil 0.5μg/kg, iv, 5min prior to reperfusion), and group D (Alprostadil 1μg/kg, iv, 5 min prior to reperfusion). The changes of hemodynamics parameter and serum cardiac tropinin I(cTnI) were detected. The pathological changes of myocardium were observed with microscope and transmission electron microscope. The hearts were removed for determination of Bcl-2 and Bax protein expression by immunohistochemistry method.Results: In group B, cTnI increased significantly at T2 and T4 compared with that at T1. At T4 , cTnI in group C and D were lower than that in group B, cTnI in group D was lower than that in group C markedly. The expression of Bax,Bcl-2 and Bcl-2/ Bax in group B,C and D are significantly different from group A. The expression of Bcl-2 protein and Bcl-2/ Bax in group C and D was increased obviously compared to that in group B, but Bax was decreased. As compared with C, Bcl-2 protein expression and Bcl-2/ Bax in group D was markedly higher and expression of Bax protein was lower.Conclusion:Alprostadil postconditioning can inhibit cTnI, Bax protein expression and increase expression of Bcl-2 protein significantly. Alprostadil can protect the mycardial from ischemia-referfusion injury.
