This paper investigates the effects of Fuscoporia obliqua polysaccharides(FOPS) on growth inhibition and apoptosis of hepatocellular carcinoma HepG-2 cells.The inhibition of cell growth was determined by MTT method.Morphological changes of the apoptosis of HepG-2 cells were observed by inverted light and transmission electronic microscope.Agarose gel electrophoresis analysis revealed DNA cleavages.Flow cytometry was used to detect cell cycle distribution of HepG-2 cells and apoptotic rate.The mRNA expression levels of the apoptosis-related gene such as Bcl-2 and Bax were detected by RT-PCR.We found that FOPS inhibited HepG-2 cells growth in vitro.The MTT test showed that FOPS could inhibit the growth of HepG-2 cells in a dosage and time dependent manner.The HepG-2 cells treated with FOPS for 48h were observed under inverted light,cell growth was sparse,clarity of kytoplasm was decreased and dead cells were increased.Transmission electronic microscope showed that cell presented some morphologic features of apoptosis, including nuclear condensation,plasma membrane blebbing and formation of apoptotic bodies. Agarose gel electrophoresis analysis showed DNA ladder.FCM assay indicated that apoptotic rate increased significantly in dose dependent manner in experimental group,and the cells were inhibited and arrested in S phase.The expression of Bcl-2 was not changed,but the expression of Bax was downregulated.FOPS inhibit the proliferation of HepG-2 cells and induce apoptosis,which may be associate with up-regulating of the expression of Bax.
