The correlative medicine of Cerebroprotein hydrolysate is biochemistry medicine which is widely used in treating the brain functionality barrier class disease's at domestic and foreign.Polyethylene glycol and the derivatives generally modify protein multi-peptide class medicine.It's widely applied in the biochemistry and the medicine domain because of its good solubility and the biological compatibility which could enhance medicine drug efficacy and extension half-life.The cerebroprotein hydrolysate are modified by the single methoxy polyethylene glycol propylaldehyde(mPEG-ALD,5K)which is preliminary researched in this thesis.The mPEG-ALD-cerebroprotein hydrolysate is obtained by separating and purifiying the modified products through modification condition's screening experiment,SDS-PAGE and HPLC.The function and influence of cerebroprotein hydrolysate and mPEG-ALD-cerebroprotein hydrolysate on newborn rats' cerebral neurocyte by vitro culture method are obersved.Measuring the length of cellular process and the diameter of cells and making contrastive analysis.Through optimized screening:pH6.0,1:1 for the mass ratio of cerebroprotein hydrolysate and mPEG-ALD,10℃,15 minutes were elected as the optimal reaction condition of modification.In this condition,the ratio could reach to 72.305%.The products which purifyed by HPLC are consistent with the SDS-PAGE,there are four kinds of priducts which molecular weight are respectively 5566.09D,5528.09D,5444.87D,5302.89D.The vitro raise's result showed that:the cerebroprotein hydrolysate and mPEG-ALD-cerebroprotein hydrolysate all have the promoter action in 5 days,but to the 7th day,only then the mPEG-ALD -cerebroprotein hydrolysate still promote the cell growth which showing that the biological activity of mPEG-ALD-cerebroprotein hydrolysate's is stable,and the half-life is prolonged.
