Cotoneaster multiflors is a kind of omamental plant widely distributed in China.And the pulp of Cotoneaster multiflors is packed with polyphenol.We study the in vitro antioxidant activity of the extracts from the pulp,stems and leaves of Cotoneaster multiflorus,the measurement of polyphenol,the extraction and the purification of the pule.Finally we established a produce process of polyphenol from Cotoneaster multiflorus pule.At the same time,we analys the seed oil of Cotoneaster multiflorus.The main conclusions as follow:1.We study the in vitro antioxidant activity of the extracts from the pulp,stems and leaves of Cotoneaster multiflorus:Basis on the result of DPPH free radical-scavenging power assay, ABTS radical cation decolorization assay,reducing power assay and Ferric reducing antioxidant power(FRAP) assay,the extracts of all those three parts of Cotoneaster multiflorus had excellent antioxidant activities.The ability of decreased in the following order:pule,stems and leaves.And the antioxidant activity of pule is obviously higher then stems and leaves,a little lower then TBHP,and higher then other synthetic antioxidants,such as VC and BHT.2.Establishe the optimum conditions for quantitative determination of total phenolic acids in Cotoneaster multiflorus pule:the volume of Folin-Ciocalteu reagent 1.0 mL,ratio between the volume of Folin-Ciocalteu reagent and 10%Na2CO3 1:2.5(v:v),reaction time 2h,reaction temperature 25℃,the detection wavelength is 765 nm.The average recovery rate was 98.72 %.It had a good accuracy and precision to determine the contents of polyphenol.3.Establishe the homogenate extraction optimum conditions of Cotoneaster multiflorus pule:Based on the Single-factor experiment,the Box-Behnken experiment design was applied to optimize the single factors of the extraction process.The optimized conditions were as follows:80%of the alcohol content,1:20 of the ratio of solid to liquid,30s of the homogenate time,and twice of the extraction.Under these conditions,the extracting ratio and purity of polyphenol from Cotoneaster multiflorus fruits were 0.82%and 14.77%respectively.4.Establishe the purification technique of Cotoneaster multiflorus pule:ues the optimumed conditions to extract the polyphenol from Cotoneaster multiflorus pule,then reextract with ethyl acetate,the extracting ratio and purity ofpolyphenol were 0.68%and 28.76%respectively.Sixteen different macroporous adsorption resins were studied,and finaly choose HPD-200A as the macroporous adsorption resin to purify the polyphenol.The conditions of static adsorption and desorption were:saturated absorption time was 4 h,the adsorption timeprature is 25℃,the concentration of solid extractant was 2 mg/mL,desorption agent was 95%(volume fraction) ethanol solution and the desorption time was 3 h.The conditions of dynamic adsorption and desorption were:the flow rate was 2 mL/min,the flow volume was 9BV,the concentration of polyphenol was 2 mg/mL,desorption agent was 95% (volume fraction) ethanol solution whose volume was 2BV at the flow rate 1 mL/min.When the content of solid extractant was 2g,we got 0.46g refined polyphenol whose purity is 72.4%.5.Analysis of oil from cotoneaster multiflorus seeds:take the method of gas chromatography-mass spectrum(GC-MS) to analysis the compounds of the oil from cotoneaster multiflorus seeds by the frist time.36 compounds and their content were identified and the identified compounds represent 99.78%of total GC peak areas.The total unsaturated fatty acid accounted for 82.85%of fatty acid content.Thus elucidate that seed oil of Cotoneaster multiflorus is a kind of functional type grease with important tnutrient,health,and medical value.
