The areca of the south drug was studied its polyphenol extraction process was researched by response surface analysis. The components of the polyphenol were determined through capillary electrophoresis method. The antioxidant activity of areca polyphenols extracts was measured. What’s more, the polyphenols extracts was purified through polyamide resin and the anti-fatigue effect of areca polyphenols on mice was tested. The purpose of this work was to provide the theoretical basis for the development and utilization of areca industry. Specific research and research results were as follows:(1) Through single factor experiments and response surface experiment, the optimum conditions which determined for the extraction of betel nut polyphenol was:the ratio of material to liquid was 1:20; the extracting temperature was 53.54℃; the extracting time was 38.41 min and the ethanol concentration was 41.32%(V/V). The total phenol content of areca husk extract was determined with the Folin-Ciocalteu method and the maximum overall yield of polyphenol was 14.96906mg/g betel nut.(2) In order to determine the components of areca polyphenols, the areca nut polyphenol extract in methanol solution was used as the research object and the method of capillary electrophoresis was studied. According to the results, six areca polyphenols were identified, the result of analysis was:(-)-epicatechin; (+)-catechin; naringenin; kaempferol; ferulic acid and chlorogenic acid.(3) The areca nut polyphenol extract in methanol solution was used as the experimental material, the DPPH radical, ABTS radical scavenging activity and reducing power to Fe3+ were chose and with BHT as the control to evaluate the antioxidant activity of areca polyphenols. The results showed that the sample had significant antioxidant effect. Although the antioxidant effect of sample was weaker than BHT, the DPPH radical and ABTS radical scavenging activity and reducing power to Fe3+ increased markedly along with the concentration of sample increasing.(4) The areca nut polyphenol extract was condensed by rotary evaporating and purified through polyamide resin.40%~80% ethanol were used to elute the areca nut polyphenol which we expected. The eluant of 75% ethanol was collected and further it was concentrated, what’s more, the purification of it was detected and the result showed that the purity was 80%.(5) The anti-fatigue effect of crude purified polyphenols of areca was studied and five conventional indicators were adopted to evaluate the anti-fatigue effect of polyphenol in mice. The mice were divided into 4 groups, namely control, low (5mg/kg body weight), medium (10mg/kg body weight) and high (20mg/kg body weight) dose groups. The swimming endurance time of mice, blood lactate concentration, blood urea nitrogen concentration, muscle glycogen content and liver glycogen content were tested and results showed that betel nut polyphenols someway had anti-fatigue effect.This work was supported by grants from the Key Technology.
