Part One: Establishing model of vestibular compensation of mimetic aging animal in ratObjective To explore the establishment of the aging vertigo effect in the vestibular compensation of mimetic aging rat.Methods Group A(D-galactose aging group, n=10), 3 months male SD rats were randomly toke 10, and treated with hypodermic 5%D-galactose(150mg.kg-1.d-1)for 8 weeks and then with intraperitoneal saline for 10 days; group B(control group,n=10)were given saline only; group C(aging group,n=10),24 months male SD rats, about weighting 450g. Weighting 1time every 7days, and dissect group A,B,C; ultraviolet spectrophotometry was used to analyze the activity of MAO2B, and Colorimetry was used to analyze the activity of SOD and LPO.Results the activity of SOD group A and C were cut down obvious(p<0.01), and compared to group A and C, the activity without significant deviation(p>0.05). the activity of LPO group A and C were cut down obvious(p<0.01), and compared to group A and C, the activity without significant deviation(p>0.05).Conclusion the mimetic aging effect in the brain of rat can be induced by D-galactose, and establishing this model can be used to study the pathogenesy of the vestibular vertigo with aging Part Two: Character of GABAB2 receptor during mimetic aging: focus on vestibular compensationObjective In the animal model of vestibular compensation, to investigate the expression of theγ-aminobutyric acid B2 receptor in the MVN of aging rat in the acute compensational time.Method In the paraffin slices of rat, using SABC immunhistochemistry technology, expression of GABAB2 receptor in the MVN of rat was examined. Then, we will analysis cell-dyeing density by computer image system, and to calculate average grey scale value x±s in the area of MVN, then do ANOVA as statistics test.Result Many neuronal cell bodies with dense GABAB2 receptor immunoreactivity were observed tube localized in the MVN of rat: compare adult rat with aging rat, we have not found obviously diversity when did the x±?s test ANOVA, then we compare the shamed adult rat with vestibular compensation rat group, the immunity response intensity of the latter is higher, p<0.05 and its result has statistical significance. Meanwhile, compared aging shamed group with vestibular compensation, its result has statistical significance. Finally, in the animal model of vestibular compensation, the positive expression is accumulating with the increasing of time in the adult and the aging rat, however, the latter's expression is obvious higher than the former.Conclusion In the aging animal models of vestibular compensation, the expression of GABAB2 receptor immunoreactivity in the MVC was effected by aging...
