Objective To investigate the effect of 2-deoxy-D-glucose on the proliferation, cell cycle and apoptosis in osteosarcoma during the normal and hypoxia environment.Methods Hypoxia culture model in vitro was established. The Cell Counting Kit-8 assay was used to examine the antiproliferative effect of 2-deoxy-D-glucose; Fluorescence microscopy was performed to observe the morphologic changes of cells; Flow cytometry (FCM) was used to examine the effects of 2-deoxy-D-glucose on the apoptosis and cell cycle of LM8 cells and DNA damage were studied through the comet assay.Results 2-deoxy-D-glucose can inhibit significantly the growth of LM8 in a concentration-dependent manner.The inhibitory effect was elevated significantly during hypoxia (P < 0.05), which was found by the higher percentage of apoptosis, arrest the cell in G2/M and S phases and cell DNA damage.Conclusion 2-deoxy-D-glucose can inhibit LM8 cell growth via arrest of cell cycle,elevated apoptosis and necrosis induction during hypoxia.
