Objectives:l.to study the effect of SafflorYellow(SY) on apoptosis of neuronal and the expression of apurinic/apyrimidinic endonuclease/redox factor 1(APE/Ref),and the neuronal apoptosis of the hippocampal CA1 subcgion in neonatal rate with hypoxia-ischemic brain damage(HIBD).2.To explore the relationship between APE/Refl and neuronal apoptosis in the hippocampal CA1 subregion after hypoxiaischemic brain damage in rats.3.To investigate the mechanism of SY neuronal protective effect after HIBDMethods:Seventy-two 7-day postnatal Sprague-Dawley(SD) rats were randomly divided into 3 groups:(1) control group(n=24),(2) hypoxieischemic brain damage group(n=24),(3) SY-treated group(n=24).In the SY-group,rats received SY at Oh and 4h after HIBD.The control group was not administrated.The expression of APE/Refl was measured by Immunohistochemical method.The apoptosis of the hippocampal cell was detected by terminal deoxymcleotidyltransferase-mediated 2-deoxyuridine 5-triphosphate-biotin nick end labeling(TUNEL) straining.The quantities of APE/Ref1 and apoptosis cells were counted.All data were analyzed by SPSS 13.0 software.R0.05 was considered statistically significant.Results:1.results of neuronal apoptosis detected by TUNEL:(1) artifical operation group: Very few number of TUNEL-positive neonatal were found by TUNEL staining in the hippocampal CA1 subregion in neonatal rats(P>0.005).(2) HIBD group:the number of UNEL-positive neonatal was markedly increased comparing with artifical operation group(P<0.005) at 12h,24h and 48h.(3) SY group:the number of UNEL-positive neonatal was reduced(P>0.005) at 6h and significantly reduced(P< 0.005) at 12h,24h and 48h comparing with HIBD group. 2.results of the expression of APE/Refl measured by Immunohistochemical method(1) artifical operation group:the expression of APE/Refl were found by TUNEL staining in the hippocampal CA1 subregion in neonatal rats(p>0.005).(2) HIBD group:the number of UNEL-positive neonatal expressed by APE/Refl was significantly reduced comparing with artifical operation group(P<0.005).(3) SY group:the expression of APE/Ref-1 was reduced at 6h comparing with HIBD group(P>0.005).The expression of APE/Ref-1 was significantly reduced comparing with HIBD group(P<0.005).3.the number of neuronal apoptosis of the hippocampal CA1 subregion was growing as the number of UNEL-positive neonatal expressed by APE/Refl.Conclusions:l.SY have neuronal protective effect on the neonatal rats after HIBD.2.The activation of APE/Ref—1 play an important role in the inhibitation of neuronal apoptosis on neonatal rats after HIBD.3.The protective mechanism of SY may be restrain the reduce/ of APE/Ref-1 and reduce neuronal apoptosis.
