Study On Gene Polymorphism Of Aedes Albopictus RDNA ITS2 And Analysis On Logistic Regression Of Epidemic Factors Of Dengue Fever

【Objective】To study gene sequences of Aedes albopictus and the epidemic factors of dengue fever in Fujian,investigate gene polymorphism of Aedes albopictus rDNA ITS2 of diffent geography ,further know the risk factors of dengue fever and provide base for prevention and cure of dengue fever.【Methods】Gather separately Aedes albopictus larvae of five spots in Fuding,Wuyishan,Hanjiang and Dongshan of Fujian,raise in Lab,collect grown-up mosquitos and conserve in -20℃with 75% alcohol. PCR and sequences analysis Aedes albopictus rDNA ITS2, analysis gene polymorphism of Aedes albopictus rDNA ITS2 in bioinformation system. Choose 35 villages of Fujian for investigations,design the investigate forms of epidemic factors of dengue fever. Analysis on logistic regression of types of geography ,density of population, density of Aedes albopictus larvae, usage of preventing mosquito facilities, fluid population to and from Southeast Asia area and imported dengue fever cases.【Results】1. PCR Aedes albopictus rDNA ITS2 of four different areas in Fujian.Gain about 500bp fragment in agar gel electrophoresis . Aedes albopictus of different areas in Fujian show Sequences analysis gene polymorphism through sequences analysis.There are divertation, transversion and deletion in rDNA ITS2 mononucleotide.However, there is not individual variation between five Aedes albopictus of the same spot.2. Analysis on logistic regression of epidemic factors of dengue fever,Aedes albopictus larvae, imported dengue fever cases and fluid population to and from Southeast Asia area are elected in regression equation,the relative risks are 7.25,9.83 and 5.64 .【Conclusions】Aedes albopictus of different areas show gene polymorphism in rDNA ITS2,which is related to longitude,latitude,altitude,temperature, rainfalland humidity. Dengue fever often happens where there are fluid populations to and from Southeast Asia area , imported dengue fever cases and high density of Aedes albopictus larvae,which offers base for the scientific prevention and cure of dengue fever.
