Objective: To investigate the impossible association and roles of APOA5 gene with Hypertensive disorders in pregnancy (PIH).Methods: Firstly, we examined the distribution of 3 known SNPs (-12238 T>C,S19W,185G in APOA5) in 149 Chinese PIH patients, 105 controls by PCR-RFLP. Finally, we analysed the association of these SNPs with lipid metabolism in patients with PIH.Results :1. There were no significant difference in age and pregancy week composition between the patients and normal pregnants(P>0.05); the frequencies of different genotypes of the 3 known SNPs (-12238,S19W,185G in APOA5) were in agreement with Hardy-Weinberg equilibrium in our study(P<0.05).2. In the normal pregnants: The frequencies of the -12238bp TC,CC,TT genotypes were 52.4%,28.6%,19%;allele frequencies were T 45.2%,C 54.8%;the frequencies of the S19W , SW,SS genotypes were30.5%,68.6%;allele frequencies were S 83.8%,W 16.2%;the frequencies of the 185 G>T ,GG,GT genotypes were 90.5%,8.6%;allele frequencies were G 94.8%,T 5.2%.3. In the patient group: The frequencies of the -12238 ,TC,CC,TT genotypes were 50.3%,31.5%,18.1%;allele frequencies were T 43.3%,C56.7%;in the frequencies of the S19W, SW,SS genotypes were 25.5%,74.5%;allele frequencies were S 87.2%,W12.8%;but with the disease degree, there were significant difference in genotypes and allele frequencies in the S19W; the frequencies of the 185 G>T, GG,GT genotypes were 87.2%,12.1%;allele frequencies were G 93.3%,T 6.7%. 4. There were no significant difference in genotypes and allele frequencies in the two groups with three SNPs ( P>0.05).5. Haplotype analyses with EH program demonstrated that the haplotypes frequencies containing 12238T/C and G185T were no significantly different between PIH and control groups ( P >0.05 ).6. There were significant difference in.the TG,TC and APOA1 levels of patients with normal group.Conclusion: There might be an association between APOA5 with Hypertensive disorders in pregnancy.
