Abstract/Summary:
[objective]: To explore the expression of Tiaml and CD44V3 in human Breast Carcinoma and to estimate the relationships of Tiaml and CD44V3 levels with clinical and pathological characteristics.[methods]: semi-quantitative no-biotin two-step immunohistochemistry staining method was applied to detect the expression of Tiaml,CD44V3,ER,PR,C-erbB-2 of 80 cases of primary invasive Breast Cancer tissues ,13 cases of breast benign hyperplastic tissues as control.[results]: The positive rate of expression of Tiaml, CD44v3 in Breast Cancer tissues showed higher than in benign hyperplastic tissues(P<0.05). The positive rate of Tamil expression in Breast Carcinoma tissues with axillary lymph node metastasis was higher than that of without (P<0.005). Significant difference of Tiaml expression was existed between the group of stages III+IV and the group of stages I+II(P<0.005). No significant difference was founded in the expression of Tiaml among age,histological types or grades,tumor size,location of tumor,ER,PR,C-erbB-2 status, expression of CD44V3 had the same result (P>0.05). CD44V3 was mainly expressed in the myoepithelial cells in benign mammary leisions,while positive staining was found on tumor cell membranes in Breast Carcinoma tissues. Expression of CD44V3 has no correlation with the axillary lymph node status(P>0.05),but it has positively statistic difference if there was papillary or basal invasion (P<0.01).In univariate analysis the expression of CD44V3 was not associated with recurrence or distant metastasis while Tiaml over expression showed a contrary result. An Wilcoxon signed-rank test between Tiaml and CD44V3 expression suggested statistical significance (T=2101.5,u=5.385,P<0.001), Then an Spearman positive correlation was shown between them in Breast Carcinoma( rs'=0.284, P<0.02). The expression of Tiam1 was showed to be significantly related to worse outcome (P<0.01). Furthermore, subgroup analysis revealed a sequentially rising recurrence or distant metastasis or death with the over-expression of Tiaml (Fisher's Exact Test: P=0.001). In Logistic analysis demonstrated Tiaml was an important role of worse clinical outcome.[conclusion]: 1.Expression of Tiaml,CD44V3 were higher in human primary Breast Carcinoma than in benign hyperplastic tissues. 2. Expression of Tiaml was closely correlated to axillary lymph node metastasis and clinical stages. 3. CD44V3 was contributed to invasive ability of the tumor tissues. 4. Tiaml plays an important role in the invasion and distant metastasis of mammary cancer. It may be an independent marker for metastasis of invasive Breast cancer. 5. combined study of expression of Tiaml,CD44V3 has practical value in prognosis of the primary Breast Carcinoma.
