OBJECT: (1)to analyze the distribution of TT Virus Genomic Groups 1-5 inFujian Province voluntary Blood Donors, HBV-infected Pataients,and HCV Carriers.(2)to analyze the Distribution of TT Virus Genomic Groups in Blood Donors, HBV-infected Pataients,and HCV Carriers.METHOD Prepare the blood serum from 106 voluntary Blood Donors,64 HBV-infected Pataients and 32 HCV Carriers.Design 24 group-specific primers.Then it was use the untranslated region PCR system (UTR-PCR) to detect the blood serum TTV-DNA in blood donors, HBV-infected Patients,and HCV Carriers. Five nested PCR assays(G1-G5PCR) were developed in an attempt to amplify specifically TTV DNAs of each genomic group(1–5). Statistic methods:it was useed the Statistic software SPSS to analyze the results detected.RESULTS:(1)The TTV-DNA infection rate in use of improvemont UTR-PCR assay was:95.3%(105/108) in voluntary blood donors,95.3%(61/64) in HBV-infected patients,96.9%(31/32) in HCV carriers.The total infection rate in the three experimental units is 95.5%。.(2)TTV genomic group 4 was found most frequently 52.0%(105/202)samples were positive), followed by group 5 (79/202,39.1%), group3(66/202,32.7%),group 1 (52/202,25.7%), and group 2 (17/202,8.4%). All genomic groups were detected in at least one blood donor, one HBV patients and one HCV carriers. Using specific PCR assays, TTV DNA average infection rate was 23.8% in blood donors, 37.8% in HBV patients, and 45.0% in HCV carriers..(3)There have 4 samples from blood donors , 3 samples from HBV patients and 1 sample from HCV carrier showed negative results by use the UTR-PCR assay,but positive resluts by use the group specific PCR assay.4 samples showed postive results by use the UTR-PCR assay,but negative results by use the group specific PCR assay. Among the blood donors, 73.6%(78/106) were infected with a unique TTV genomic group ,the 19.8%(21/106) others were coinfected with TTVs from 2 genomic groups.Among the HBV-infected patients, 29.7%(19/64) were infected with a unique TTV genomic group, 56.3%(36/64)were coinfected TTVs with 2 genomic groups.Among the HCV carriers, 28.1%(9/32)and31.2%(10/32) were coinfected TTVs with1 and 2 genomic groups.(4) Nucleotide sequences of G1-G5PCR products(2 positive product in each group) showed high homology with a least one of the TTV isolatesvbelonging to their presumed groups,and lower (22-71%) homology with the other groups.CONCLUSION: It was found the TTV infected is very frequently in Fujian Province voluntary Blood Donors,HBV-infected patients and HCV carriers.Using the group-specific PCR assay,all the 1-5 groups TTV could be dected from three experimental units.The TTV infection rate in HBV-infected patients and HCV carriers was high than in voluntary blood donor units.TTV can coinfected with difference genomic groups. Nucleotide sequences of PCR products were compare with those of 15 reference isolates (three of each genomic group, 1–5),The results suggest the group-specific PCR assay is a successful method.
