| Objective:To study the histocompatibility of the nanophase hydroxyapatite material on SD rats spongy bone.Evaluated the growth of the osteoplasts and the expression of inflammatory cytokines in vitro.Methods:0.01%nanophase hydroxyapatite particles and general hydroxyapatite particles were added into the osteoplast culture medium of SD rat respectively.Collect the supernatant on 1,7 and 14 days to detect the expressions of cytokines by ELISA,such as IL-1β,IL-6 and TNF-α.Seed 3×10~4/ml osteoplasts on the top of nanophase hydroxyapatite wafers and the general hydroxyapatite wafers in 24-well plate.Incubated 1,7,14days and collected the wafers.Results:The expressions of IL-1β,IL-6 and TNF-αin nanophase HAP particles were lower than the general Hydroxyapatite particles.The outcomes of IL-1β,IL-6 and TNF-αin HAP group raised significantly with the culture time prolonging.The outcomes of three indexes in Nano-HAP group did not raise obviously.The levels of IL-β,IL-6 and TNF-αpeaked on 14 days.The Expressions of IL-1βand IL-6 in Nano-HAP groups are lower than that of HAP groups on 1 and 7days(P<0.05).And the levels of them in Nano-HAP groups are significant decreased than that of HAP groups on 14 days(P<0.01 ).The outcome of TNF-αin Nano-HAP group was less than that of HAP group on the 3 timepoints.But there was no difference on 1 day and statistics difference on 7 and 14 days(P<0.05).The extracellular matrix of HAP osteoplasts are more than the general Hydroxyapatite wafers in electron microscope. There are some mineralize nodus on the surface of HAP osteoplasts.Conclusion:1.Compared with the general hydroxyapatite material,the Nanophase Hydroxypatite material is favorable for the growth and mineralization of osteoplasts in vitro.2.Compared with the general hydroxyapatite material,the expressions of TNF-α, IL-1β,IL-6 in HAP particles were lower than the general Hydroxyapatite particles. It can decrease the incidence rate of osteolysis.3.Compared with the general hydroxyapatite material,the Nanophase Hydroxypatite material has a good histocompatibility in the application. |
