ObjectiveTo propose the new knowledge about seizure etiology,and establis new treatments.To observe antiepileptic effect of the expectorant and benefiting vital energy method to pilocarpine-induced epileptic rats,and the comprehensive regulation to hippocampal neuronal apoptosis of them.To explore the pathogenesis of epilepsy and possible mechanism restrain epilepsy of the experimental drug(expectorant 1 prescription).To establish foundation for drug development and provide theoretical basis for clinical treatment.MethodsReference to ancient and modern literature epilepsy summary.The use of lithium chloride-intraperitoneal injection of pilocarpine Ways set up temporal lobe epilepsy model in rats,Intraperitoneal injection of the group before the method chosen to give the corresponding drug administered orally.Garage for 7 days,PILO group set up model and the intervention group on the 1st Huatan model,To the control group with normal saline instead of pilocarpine injection;Immunohistochemistry of rat hippocampus were observed tumor necrosis factor-a(TNF-a)and glial fibrillary acidic protein(GFAP) positive cells in the changes of expression. Experimental study consists of two parts;No.1 by measuring Huatan epileptic rats to study changes in behavior,seizure latency, duration,Discussion on the 1st Huatan the role of anti-epileptic and epileptic rats acts to improve the study and other mechanisms; Discussion on the 1st of Huatan pilocarpine-induced seizure in rat hippocampus TNF-αand glial fibrillary acidic protein (GFAP)positive cells.Results1.Behavioral observation:normal saline control group of rats without any seizures.PILO model group has 80.00%(24/30) of the animals over Racine seizure classification classⅢ;10.00%(3/30) 10.00 of the rats died;10.00%(3/30)of the rat classⅢnot attack; Animal spontaneous seizures(SRMS) for 4.51±1.48 times/only. Huatan intervention on the 1st classⅢrats attack happened more than 76.70 percent rate(23/30);Mortality rate was 6.67%(2/30); 16.67%(5/30) of the rat classⅢnot attack;SRMS was only 2.81±1.23 times/only.2.Histopathological observation:District hippocampus in control group no significant changes in neurons.PILO epilepsy model rats 3h(SE3h),hippocampal hilar,CA1,CA3 areas scattered in the beginning neuron degeneration and necrosis,cells with irregular,a small number of nerve cell loss;To SE3d,the above-mentioned pathological changes of the peak,dentate gyrus granule cell degeneration also began to appear;SE14d when damaged neurons gradually restore the situation,the cells gradually neatly arranged.1 Huatan PILO-intervention group and model group,the nerve cells at different time points significantly reduce injuries.3.Immunohistochemistry Results:Saline control group,TNF-a in rat brain tissue was stained light cytoplasm,few have nuclear immunoreactivity;and PILO in model group,TNF-a immunoreactive products are mainly in the cytoplasm,the particles were fully mechanized yellow expression,The number of positive cells began to increase when SE3h to CA1,CA3 and dentate gyrus of the main door area,SE3d and peaked in,and then gradually decline to SE28d still slightly higher than the normal level;Huatan 1 intervention group at different time points,TNF-a cytoplasmic expression PILO group were decreased significantly.Saline control group,only a small number of GFAP-positive cells,cell body,small,small,less protruding;PILO model group,GFAP-positive cells increased cell body cross-sectional area,number of processes and their branches, and the changes of ceils with TNF-a-positive cells of similar; Huatan 1 intervention group at different time points the expression of GFAP-positive group was significantly weaker than those of PILO.ConclusionLithium chloride-pilocarpine model of chronic epilepsy can replicate the human temporal lobe epilepsy with similar changes in behavior and pathology.Astrocyte proliferation in hippocampus at the early onset of epilepsy seizures and neuronal damage of the adaptive response,which may promote the late recurrence of epilepsy.Huatan 1 star hippocampus can inhibit the proliferation of glial cells.At the same time,the impact of epilepsy in rats changes in behavior,attack latency,such as changes in pathology. Prompted its possible there is inhibition of seizures.
