ObjectiveGlioma has the highest morbidity among intracranial tumors,invasive metastasis is the main reason which causes failure of treatment and death,and it is closely related to the changes of multiple genetic and molecular levels.So far clinical results are still unsatisfactory.Surgery only can excise the visible tumor tissue,and can not remove the causes of tumor incidence,so recurrence can not be avoided.From the molecular level in recent years,awareness and treatment of glioma has become a hot area of neural tumor.With the development of molecular biochemistry and the continuous development of genetic science,we have found many cell factors.The cytokines for the diagnosis and treatment of glioma has provided a new direction. This research contrasts the expressions of CTGF and TGF-β1to the pathologic characteristic and grading of human glioma,evalusion relations between CTGF and TGF-β1 and pathological grades of tumor,and discussion their effect on formation and development of glioma.MethodsSpecimen:49 cases human brain glioma specimen which cut off by surgery in neurosurgery department China Medical University affiliated NO.1 hospital from January 2006 to December 2006.Male 29 cases and female 20 cases age are from 13 to 70 years old.All the cases didn't accept radio-or chemical-therapy before surgery.The pathological granding was classified according to the standard pathological grades of World Health Organization(WHO) in 2000 for glioma GradeⅠ9 cases,GradeⅡ12 cases,GradeⅢ17 cases,GradeⅣ11 cases.Mainly agent:CTGF anti-human monoclonal antibody of the rabbit bought from Wuhan BOSTER Company.TGF-β1 anti-human multiclonal antibody of the rabbit,SP staining kit and DAB kit both bought from Beijing Zhongshang Company. Empirical method and result assessment:using SP immunohistochemical method to detect the expression information of CTGF and TGF-β1 in human brain glioma.We took one group to dye as HE to verify the pathological grades of glioma and then we used SP to detect the expression of CTGF and TGF-β1.Immunohistochemical positive is defined as brown drops in cytoplasm.Count the positive cells less than 10%(+), positive cells 10%-50%(++),positive cells more than 50%(+++). Statistical analysis:We combined gradeⅠand gradeⅡof glioma to low grade glioma (LGG),and gradeⅢand gradeⅣto high grade glioma(HGG).All data usesx~2 test.α=0.05.ResultsCTGF positive staining mainly locates in cytoplasm,like buffy or brown fine grain,sporadic weak-positive express in low grade glioma(HGG) expresses rate is 28.57%(6/21).However,expression is obviously enhanced in high grade glioma (HGG),expression rate is 85.71(24/28).There is significant deviation between two groups(P<0.05).TGF-β1 positive staining mainly locates in cytoplasm,brown fine grain like, sporadic weak-positive expresses in low grade glioma(LGG),expression rate is 33.33%(7/21).However,expression is obviously enhanced in high grade glioma(HGG),expression rate is 71.43%(20/28).There is significant deviation between two groups(P<0.05).The positive correlation was found between the score of TGF-β1 and that of CTGF.ConclusionsThe expression of CTGF and TGF-β1 had positive correlation with the pathological grades of glioma.The positive correlation was found between the score of TGF-β1 and that of CTGF.
