PurposeIntracranial glioma is common malignant tumor, which has strongly invasive power and recurrence fast. It is one of the most formidable of refractory tumors in neurosurgery, so far clinical results are still unsatisfactory. Treatment is the difference in the impact of its important factor, but the tumor inherent biological characteristics, in particular the difference of the molecular level should be the key element. The growth, metastasis and prognosis of invasive malignant tumor depend on angiogenic. The rich blood supply of tumors are often associated with the degree of malignant behavior. And the formation of tumor blood vessels is a very complex process and regulated by many factors. This research contrasts the expressions of VEGF and CTGF to the pathologic characteristic and grading of human glioma, evalusion relations between VEGF and CTGF and pathological grades of tumor, and discussion their effect on formation and development of glioma.MethodsSpecimen: 49 cases human brain glioma specimen which cut off by surgery in neurosurgery department China Medical University affiliated NO.1 hostipal from January 2006 to December 2006. Male 29 cases and female 20 cases age are from 13 to 70 years old. All the cases didn't accept radio-or chemical-therapy before surgery. The pathological granding was classified according to the standard pathological grades of World Health Organization(WHO) in 2000 for glioma.Grade I 9 cases, Grade II 12 cases, Grade III 17 cases and Grade IV 11 cases.Mainly agent: CTGF anti-human monoclonal antibody of the rabbit bought from Wuhan BOSTER Company. VEGF anti-human multiclonal antibody of the rabbit, SP staining kit and DAB kit both bought from Beijing Zhongshang Company.Empirical method and result assessment: using SP immunohistochemical method to detect the expression information of VEGF and CTGF in human brain glioma. We took one group to dye as HE to verify the pathological grades of glioma and then we used SP to detect the expressions of VEGF and CTGF. Immunohistochemical positive is defined as brown drops in cytoplasm. Count the positive cells less than 10%(+), positive cells 10%~50%(++), positive cells more than 50%(+++).Statistical analysis: We combined grade I and grade II of glioma to low grade glioma(LGG), and grade III and grade IV to high grade glioma(HGG). All data uses X~2 test.
