| ObjectiveLung carcinoma is one of the most common malignancies of the carcinoma and greatly threatens human health and life.The mechanism of tumorigenesis remains unclear for Lung carcinoma.Most Lung carcinoma patients are already in the late stage at the time of diagnosis due to lack of specific symptoms,often with lymphatic metastasis,and cannot be treated effectively.Therefore,it is very important clinically to understand the role of lymphangiogenesis in the metastasis of Lung carcinoma and to search for the anti-lymphangiogenesis agents to block Lung carcinoma metastasis.In Tumor metastasis suppressor gene's family,the KISS-1 and E-cadherin both in the malignant tissue is conducive to loss of our tumor cell invasion and metastasis in many tumors has been confirmed the purpose of our experiment is to examine the KISS-1 and E-cadherin in non-small cell lung cancer the expression of the meaning and the relationship between the two.Material and method1.The source of cases80 cases of the non-small cell lung cancer tissues and 20 cases of normal lung tissues are from the department of chest surgery of ShengJing hospital of China Medical University from Mar,2005 to Feb,2008.2.MethodImmunostaining of KiSS-1 and E-cadherin in 80 cases of carcinoma tissue respectively,and 20 cases of normal tissue as control group.3.Standard of consequenceWe use the double-blind methods to observe the results,KiSS-1 protein metastin and E-cadherin to cytoplasm,cell membrane staining;in the light micro- scope,the response has brown particles in the cytoplasm,cell membranes are positive in the cells,in five different under the same range of high power field counted 500 cells and immunohistochemical staining positive cells,positive cells of less than 5%for(+),5%~50%(+),more than 50%~75%(++),greater than 75%of(+++).4.Statistics analysisSPSS13.0 is used to analysis the data.chi-square is used to analysis the difference between case and contral group in KiSS-1 and E-cadherin respectively.A pvalue of less than 0.05 was considered significant.Spearman rank correlation analysis is used to analyse the relation of KiSS-1 and E-cadherin.Result1.Expression of KiSS-1 and E-cadherin in non-small lung cancer(1) KiSS-1 in normal lung tissue in the cytoplasm,the membrane was stained brown,whereas in non-small cell lung cancer with positive expression was significantly reduced,a significant difference(P<0.05)(2) E-cadherin in the normal lung tissue in the cytoplasm,the membrane was stained brown,whereas in non-small cell lung cancer with positive expression was significantly reduced,a significant difference(P<0.05)2.Expression of KiSS-1 and E-cadherin with clinical pathology characteristic(1)In accordance with non-small cell lung cancer in different clinical stages(P<0.05),lymph node metastasis(P<0.05) different clinicopathological parameters such as,KiSS-1 positive expression rate were significantly different,but the tumor age,pathological type and differentiation.(2)Non-small cell lung cancer based on lymph node metastasis(P<0.05),pathological stage(P<0.05) and the degree of cell differentiation(P<0.05).Diffent clinic pathological parameters such as,E-cadherin positive expression rate were signicantly different,but the pathological type and age is irrelevant.3.The relationship between KiSS-1 and E-cadherinThe positive expressions of KiSS-1 and E-cadherin were associated. Conclusion1.The expression of KiSS-1 and E-cadherin in case and control group have a significant difference.2.KiSS-1's loss of expression,as well as E-cadherin,may be a biogical maker in non-small cell lung cancer.The levels of KiSS-1 were useful molecular markers for evaluating malignancy degree and lymph node metastasis of in non-small cell lung cancer.3 KiSS-1 is associated with the expression of E-cadherin. |
