| Objective:Through studying the research of NF-κB and TNF-αin MS rats by using advanced technology and methods in order to explore the function of the treatment mechanism of MS.Methods:Sixty 8-week-old mate SD rats were randomly divided into 2 groups which are vacuity group and experimental group.Vacuity group was fed with common diet,and experimental group was fed with high salt,high lipid and high glucose diet for 20 weeks to induce experimental metabolic syndrome.The 37 MS rats were randomly divided into 4 groups:the model group,medicine control group,huanglianwendan decoction high dose group, low dose group.Detection of rat body weight,serum insulin,blood glucose, blood lipids and blood pressure and other indicators;Using the RIA measured serum concentrations of TNF-αchange;immunohistochemistry determination of rat aortic NF-κB changes.Results:After 20 weeks weeding with high-fat,high-salt high glucose,MS model rats weight,abdominal circumference,blood pressure,FBG,FINS,HOMA-IR,TG,TC and LDL-C are significantly higher than the normal group, Compared with normal group are significantly different(P<0.01).The HDL-C level lower than normal group,compared with the normal group are significantly different(P<0.01).The effects of Huanglian Wendan Decoction on weight,blood presure,glycometabolism,lipid metabolism,inflammatory factor and adhesions of laboratory MS rats.Serum tests of glycometabolism showed that Huanglian Wendan Decoction can significantly lower fasting blood sugar and insulin and thus improve IR condition,similar to the effect of metformin.Serum tests of lipid metabolism showed that Huanglian Wendan Decoction can significantly lower the level of serum cholesterol,triglyceride and low-density lipoprotein and lower the level of blood pressure and the weight of MS mice.Their function in regulating lipid metabolism and in adjusting blood pressure are better of to that of metformin.High-dose group and control group serum concentrations of TNF-αwere lower,and with the model group has significant different(P<0.01).High-and low-dose group the expression of NF-κB compared with the model group were significantly lower,and high-and low-dose group between the expression of NF-κB significantly different(P<0.01).Conclusions:1.It can build MS model with feeding high-fat high-sugar, high-salt diet to the rats.2.Huanglian Wendan Decoction can loss weight,lower blood pressure,regulate lipid and improve insulin resistance,which can improve the various components of MS.3.Huanglian Wendan Decoction can reduce the serum TNF-αconcentration,improve aortic NF-κB expression to achieve the purpose of the treatment of MS,the treatment of MS is effective medicine. |
