Expression Of Human Thymic Stromal Lymphopoietin In Ovarian Cancer

Ovarian cancer is one of the most important enemies to oncologists.Ovarian cancer is hard to find early.Most cases of ovarian cancers are detected at late stages. Metastasis and relapse can be seen frequently.In most of these cases,we can find local invasion and peritoneal metastasis.Ovarian cancer has many histological subfamilies.Epithelioid ovarian cancer is the most common kind,which takes 85%-90%of malignant ovarian tumors.Much emphasis was recently given to tumor-related immunology.How does tumor induce immune tolerance and immunosuppression is unclear.Human thymic stromal lymphopoietin is one of hematopoietic growth factors.It can induce Th2 bias and promote the differentiation and development of regulatory T cells.It is also involved in pathogenesis of autoimmune diseases.In order to investigate whether TSLP exists in ovarian cancer and its possible role in tumor-immune escape,the present study made this elementary evaluation.OBJECTIVE:To ivestigate whether TSLP exists in human ovarian cancer tissues and human ovarian cancer cell line.METHODS:Immunohistology was used to analyse the expression of TSLP in normal ovary and malignant ovarian cancer.TSLP expression of SK0V₃(human serous papillary cystadenocarcinoma cell line) was analysed by means of immunocytochemistry.ELISA(enzyme linked immunosorbent assay) wais used to detect the concentration of TSLP in supernatant of SKOV₃.RESULTS:TSLP is unexpressed or low-expressed in normal ovary epithelial cells, but high-expressed in cytoplasm of serous cystadenocarcinoma and mucinous cystadenocarcinoma cells.There is significant statistic difference.TSLP is high-expressed in cytoplasm of SKOV₃ cells,but undetectable in supernatant of SKOV₃.CONCLUSIONS:TSLP is high-expressed in cytoplasm of serous and mucinous ovarian cancer cells than in normal ovary.SKOV₃ cells high-express TSLP.But SKOV₃ cells do not secrete TSLP.
