Objective:The extract of erigerion could relieve the pathological lesion of acute rejection after transplantation.This study want to investigate the preventive effects of the extract of erigeron on chronic renal allograft rejection and to explore its mechanism.Methods:1,The rat model of chronic renal allograft rejection was made with microsurgery and adopting classic model of chronic rejection after renel transplantation.F344 rat was adopt as donator and Lewis rat was adopt as receptor.The receptors were given CsA by the dose of 5mg/(kg·d) 10 days by intragastric administration to prevent the occurrence of acute rejection.2,The recipients were divided into four groups,including control group(same volume of 0.9%saline solution),immunosuppressive grop(mycophenolate mofetil, 20mg/kg) and low(5mg/kg)and large(10mg/kg) dose of the extract of erigerion groups.Recipients were given drugs every day by intraperitoneal injection from 10th day to 12th week after operation.3,Serum creatinine(Scr) and 24-hour urine protein(Upro) were measured at 4, 8,and 12 weeks after operation.To measure the density of VEGF and to examine the histology of kidney grafts on week 12 after operation.4,The income data were represented as mean±SD.We arranged the date by the EXCEL database and analysised the date by SPSS13.0 statistics software.We use the method of analysis of variance and P<0.05 indicates the difference has statistical significance.Result:1,By histology examination,the allgraft was scored according to Banff 97.The score of four groups were 9.50±0.93,8.38±0.74,7.38±0.74 and 5.75±0.71.The score of low dose group,large dose and immunosuppressive grop was low than control group(P<0.05).The score of large dose group and immunosuppressive grop was low than low dose group (P<0.05).The score of immunosuppressive grop was low than large dose group(P<0.01).2,As compared with those in the control group,levels of Scr in the treatment groups were markedly declined on week 4,8 and 12,and those in large dose group,immunosuppressive grop were obviously declined to compare with control group (P<0.01).Levels of Scr in low dose group was declined in the 8th and 12th week after operation compared with control group(P<0.05).Levels of Scr in immunosuppressive grop was markedly declined in the three time points compared with low dose group (P<0.01) and was markedly declined in the 8th and 12th week after operation compared with large dose group(P<0.05).Levels of Scr in large dose group was obviously declined to compare with low dose group(P<0.05).3,24-hour Upro of rats in immunosuppressive grop and low dose group were decreased to compare with control group(P<0.05).24-hour Upro of rats in large dose grop was decreased to compare with control group in the 8th and 12th week after operation(P<0.05).24-hour Upro of rats in immunosuppressive grop and large dose grop was decreased to compare with low dose group in the 8th and 12th week after operation(P<0.05) and large dose grop in the 12th week after operation(P<0.05).The difference of 24-hour Upro of rats in large dose grop and that in low dose grop were not statistically significant.4,At the end of the 12th week the density of VEGF were decreased in the treatment groups compared with control group and that in low and large dose group was increased compared with immunosuppressive grop.The density of VEGF in large dose group was decreased compared with low dose group.All the diference has statistical significance(P<0.01).Conelusions:1,The extract of erigeron plays a protective role in the function and structure of the kidney graft.It can delay the occurrence of chronic allograft rejection.The prevention and cure effect to chronic allograft rejection of mycophenolate is surpasser than that of the extract of erigeron.2,The extract of erigeron could protect the function and structure of transplantation may by the way of reducing the produce of VEGF.
