Objects:To compare the difference of low abundant proteins in the serum of breast cancer parients with different prognosis,with a view to find some new biomarkers to impace the prognosis of breast cancer patients.Methods:Collect the serum of breast cancer patients with different prognosis. Deplete six kinds of high abundant protein using Multiple Affinity Removal System. Collect low abundant protein and digest them with trypsin into peptides.To research the difference of peptides between the subgroups using Agilent 1100 Series Liquid Chromatography-chip / mass spectrometry system,analyze and identified the output with Agilent Spectrum Mill protein database professional statistical analysis software. Of research findings,the use of ELISA method for authentication.Statistical analysis data were processed by SPSS13.0 software.Results:1,Proteomics research findings:The expression of membrane proteins in favorable prognosis group(LN-,ER+,PR+,HER2-) are positive,and in unfavorable prognosis group(LN+,ER-,PR-,HER2+) are negative,such proteins are Mucinl3,Desmoplakin,Desmoglein-1,ADAM 17,Monocyte differentiation antigen CD 14,Relaxin receptor 1 and Tyrosine-protein kinase CSK.The expression of membrane proteins in favorable prognosis group(LN-,ER+,PR+,HER2-) are negative and in unfavorable prognosis group(LN+,ER-,PR-,HER2+) are positive,such proteins are Vitamin K-dependent protein C,Thrombospondin-1,Coagulation factor X and Coagulation factor IX.2,The concentration of serum sCD14 is significantly different between favorable prognosis group and the three subgroups of unfavorable prognosis group grouped according to the number of lymph node metastasis(l-3;4-9;≥10),P<0.05;but no significantly different between the three subgroups,P>0.05.That is the concentration of serum sCD14 have no direct relevance with the number of lymph node metastasis in breast invasive ductal carcinoma of LN+,ER-,PR-,HER2+.3,The concentration of serum sCD14 is significantly different between unfavorable prognosis group and the three subgroups of favorable prognosis group grouped according to the extent of ER-positive(≤30%,>30%,≤60%and>60%),P<0.05;but no significantly different between the three subgroups,P>0.05.That is the concentration of serum sCD14 have no direct relevance with the extent of ER-positive in breast invasive ductal carcinoma of LN-,ER+,PR+,HER2-.4,The concentration of serum sCD14 is significantly different between unfavorable prognosis group and the three subgroups of favorable prognosis group grouped according to the extent of PR-positive(≤30%,>30%,≤60%and>60%),P<0.05;but no significantly different between the three subgroups,P>0.05.That is the concentration of serum sCD14 have no direct relevance with the extent of PR-positive in breast invasive ductal carcinoma of LN-,ER+,PR+,HER2-.5,The concentration of serum sCD14 is significantly different between favorable prognosis group and the two subgroups of unfavorable prognosis group grouped according to the extent of HER2-positive(HER2++ and HER2+++),P<0.05;but no significantly different between the two subgroups,P>0.05.That is the concentration of serum sCD14 have no direct relevance with the extent of HER2-positive in breast invasive ductal carcinoma of LN+,ER,PR,HER2+.6,The expression of membrane CD 14 in monocytes / macrophages of breast cancer tissue exists in both favorable prognosis patient and unfavorable prognosis patient, but no significantly different between the two subgroups. higher sensitivity,specificity and prediction value.7,The AUC in ROC curve of the concentration of serum sCD14M and sCD14 of 80 cases of breast invasive ductal carcinoma are 0.839 and 0.799,respectively,withConclusions:1,Patients of breast invasive ductal carcinoma with high concentrations of Mucinl3,Desmoplakin,Desmoglein-1,ADAM 17,Relaxin receptor 1 and Tyrosine-protein kinase CSK in their serum may indicate better prognosis.2,Patients of breast invasive ductal carcinoma with high concentrations of Vitamin K-dependent protein C,Thrombospondin-1,Coagulation factor X and Coagulation factor IX in their serum may indicate poorer prognosis.3,The concentration of serum sCD14 have no direct relevance with the number of lymph node metastasis in breast invasive ductal carcinoma of LN +,ER-,PR-,HER2 4,The concentration of serum sCD14 have no direct relevance with the extent of ER-positive in breast invasive ductal carcinoma of LN-,ER+,PR+,HER2-.5,The concentration of serum sCD14 have no direct relevance with the extent of PR-positive in breast invasive ductal carcinoma of LN-,ER+,PR+,HER2-.6,The concentration of serum sCD14 have no direct relevance with the extent of HER2-positive in breast invasive ductal carcinoma of LN+,ER-,PR-,HER2+.7,The concentration of serum sCD14 is significantly different between favorable prognosis group(LN-,ER+,PR+,HER2-) and unfavarable prognosis group(LN+,ER-,PR-,HER2+),P<0.05.8,The expression of membrane CD 14 in monocytes / macrophages of breast cancer tissue exists in both favorable prognosis patient and unfavorable prognosis patient, but no significantly different between the two subgroups.9,The AUC in ROC curve of the concentration of serum sCD14M and sCD14 of 80 cases of breast invasive ductal carcinoma are 0.839 and 0.799,respectively,with higher sensitivity,specificity and prediction value.
