Objective To observe recipient humoral immunity and cell-mediated immunity after mice's abdominal aorta transplantation.Methods To establish a mouse model of abdominal aortic transplantation, found experimental group(allogeneic transplantation group) and the comparison group(isogeneic transplantation group),a different time period(3,7 days post-transplantation) gain recipient mouse whole blood cells,serum and arterial graft.Flow cytometry(FCM) detect the ratio changes of the blood CD4+T cells, CD8+T cells in mice.Enzyme-linked immunosorbent assay(ELISA) detect the immunoglobulin G(IgG) changes before and after surgery in recipient mice serum. Examine the transplanted Artery by histological examination.And by immunohistochemistry detect grafted artery partial deposition of IgG.Results After transplantation,Experimental group artery appear inflammatory response,till 7 days,transplant vascular segment can be seen on each layer are a large number of lymphocytic infiltration;endothelial cell swelling,loss,necrosis,focal necrosis in the Tunica media.Post-transplantation,each time Group CD4,CD8 percentage of positive cells,CD4/CD8 ratio was significantly higher than preoperative(P<0.01).After transplantation 3 days,7 days,CD4/CD8 ratio and the degree of graft rejection was positively correlated Post-transplantation,each time Group the IgG significantly higher than preoperatively(P<0.01),IgG ratio and the degree of graft rejection was positively correlated.In Immunohistochemistry,IgG can be seen in the transplant artery deposition in Tunica media.But inflammatory response in comparative group vascular was a slight.Comparative group, preoperative and postoperative each time group CD4,CD8 percentage of positive cells;CD4/CD8 ratio did not change significantly(P>0.05).Preoperative and postoperative IgG at each time group has no significant change(P>0.05).Transplant Arteries has not seen obvious positive IgG staining.Conclusion Allogeneic transplantation of abdominal aorta can induce acute rejection.Taking cd4 as a cell mediated immunity represent and IgG as a humoral immunity represent both has early contributing in acute rejection.
