Effects And Mechanism Of Five Common Chinese Herbal Medicine In Mice Immunity

In advocating green consumption today, the business outlook of antimicrobial chemicals is not optimistic: The EU had announced in 2006 that antibiotics no longer were used as additives to promote production, and banned many veterinary drugs. Application of Chinese herbal medicine to improve the immunity of animals, prevent and control animal diseases in recent years was one of the main elements. In this study, mice were studied using five common Chinese herbal medicine astragalus, dangshen, Poria, Chenpi, and hawthorn, and observated that single ingredients and the compound in vitro and in vivo were how to influence immune function, and to explore its mechanism in immune system of animals as follows:1 Immunity Test in VitroThe extracts of hawthorn from Astragalus, dangshen, Poria Chenpi, polysaccharide as single ingredient [Astragalus (H), Poria (F), dangshen (D)], two compatible compound drugs (DH, DF, FH) and five compatible compound drug (Z), the concentration from which was each set to 100,10,1,0.1μg/ml, the concentration of each was repeated 8 hole. The results suggested that: Some single Chinese herbal medicine and the compatibility both had the concentration-dependent two-way immune regulation to ConA-induced in vitro T cell proliferation in mice; Some single-compatible and compound drugs also had dose-dependent two-way immune regulatory role in vitro LPS-induced B lymphocyte proliferation in mice.2 Immunity Test in Vivo40 ICR mice with the weight of 18~22g were randomly divided into 5 groups, namely: the control group, the positive control group, dangshen Astragalus (DH) treatment group and the compound treatment group (Z). The mice were given at the fortified levels of 40,120,360 mg/kg dose of drug in oral administration for 14 days, and weight every 3 days. In 4,8 and 12 days the mice were given cyclophosphamide in the administration of intraperitoneal injection at the level of 50 mg / kg, after 14 days, separated serum from spleen and thymus, to do lymphocyte translation and test organ coefficient, change of weight, the influence of medicine on the function of celiac macrophage in rats was measured with carbon particle clearance test.The results suggested that: DH (40mg/kg) and Z(120mg/kg) can be very significant in promoting the value of T lymphocyte responses in mice (P<0.001) when the mice were immunized with OVA. DH and Z in a certain dosage can significantly improve the mice phagocytic index. DH (40mg/kg) and Z(120mg/kg) can be significantly enhanced with coefficient of the spleen (P <0.05); DH (40mg/kg), Z (120mg/kg) and the positive control group can be significantly or very significantly enhanced the thymus organ coefficient (P<0.01, P<0.05). DH (40mg/kg) can obviously increase weight of mice ,but a total dose of the compound no significant gaining in weight;25 ICR mice with the weight of 18~22g were randomly divided into 5 groups, namely:the normal control group, levamisole group (the positive control group, levamisole 50 mg / kg), dangshen Astragalus compound treatment group (DH20, 40 and 80 mg / kg), in oral administration one time on every day for 15 days. In 4,8 and 12 days the mice were given cyclophosphamide in the administration of intraperitoneal injection at the level of 50 mg / kg, before the administration of 1 and the 7th day, subcutaneous injection of OVA and aluminum adjuvant to immunize the mice, separated serum on the 8th day after immunization, to test serum antibodies. The results suggested that:DH (40mg/kg) can significantly or very significantly increase in the concentration of serum IgG, IgG1 and IgG2b antibodies (P<0.001, P<0.01 or P<0.05) when the mice were immunized with OVA.The experimental results showed that DH (40mg/kg) can promote the specific and non-specific immunization and also promote immune organs of the mice obviously, DH (40mg/kg) can enhance immune health effects of animals. The study had positive significance for Chinese herbal medicine resources applying for the development of animal health products.
