| Objective1.To study the relationship between metalloproteinases-9,complement 3 and the edema after intracerebral hemorrhage(ICH);2.To explore the effect and the mechanism of Xingnaojing on edema induced by ICH and to investigate the underlying clinical mechanisms.Methods1.Seventy-two healthy adult male Sprague-Dawley(SD) rats were randomly divided into three groups,ICH group,ICH with Xingnaojing treatment group(XNJ group) and sham-operation group.2.Saline(1ml/kg)(for sham-operation group and ICH group) and Xingnaojing injection(1ml/kg)(for XNJ group) were injected respectively through vena caudalis right after operation;3.The rats were anesthetized with intraperitoneal injection and fixed on the stereotactic frame.A microsyringe was inserted stereotaxically into the right caudate nucleus(coordinates:0.2mm anterior,5 mm ventral,and 3mm lateral to the bregma)of the rat,and 50ul auto-non-anticoagulated blood were given(XNJ group and ICH group) or nothing was given(sham-operation group);4.The neurofuction of rats was evaluated immediately and at 24,48,72 hours after operation;5.The water contents in the area of peri-hematoma were examined respectively at 72 hours after operation in every group.6.EB solution,which cycles in vivo for 2 hours,was given through vena caudalis separately at 70 hours post-operation,and then the rats were killed and the brain were removed for the detection of the contents of EB in brain tissue.7.Four percentage paraformaldehyde was injected into the heart and cycled in vivo body,then the rats were decapitated.The expression of MMP-9 and C3 around the hematoma were measured with immunohistochemistry technology.Results1.Neurobehavioral scores were decreased in the rats with intracerebral hemorrhage. As compared to the sham-operation group,neurobehavioral scores were lower in the ICH group at every time points(P<0.05).However,neurobehavioral scores increased gradually and peaked at 72h after the treatment of xingnaojing injection,.At 48h and 72h post-operation,scores of XNJ group was higher than that of ICH group (P<0.05).2.At 72h after the operation,water content of ICH group was higher than that of sham-operation group(P<0.05).The water content in brain tissue of XNJ group was lower as compared with ICH group(P<0.05).3.The permeability of blood-brain barrier increased in ICH group at 72h post-operation. The content of EB in the area of perihematoma in ICH group was higher than that of the sham-operation group(P<0.05),as well as XNJ group(P<0.05).4.The number of positive cell labeled with MMP-9 and C3 in the ICH group was higher than that of the sham-operation group(P<0.05),and the expression of MMP-9 and C3 around hematoma in XNJ group lower than in ICH group(P<0.05).5.The gedtalt form of the brain:the bilateral hemispheres were appozimately symmetric in the sham-operation group.The enlarged hemisphere,the extended gyri and the shallow sulci were observed in the right cerebral hemisphere of ICH group. While the appearance of the bilateral hemispheres of XNJ group changed slightly.6.HE staining:No obvious pathological changes were observed under Light microscope in the sham-operation group.In ICH group,the nervous tissues around hematoma were loose and inflammatory cells transmigrated into the area of peri-hematoma.,The neural ballooning degeneration and necrosis were observed in the brain tissue of ICH group.While the pathological changes in the brain tissues of XNJ group were not as serious as that of ICH group. Conclusion1.The preparation of this ICH rat model,established by stereotaxic injection of auto-non-anticoagulated blood,was relative easy,reliable and practical.2.It is proved that the breakdown of Blood-Brain barrier induces cerebral edema after incerebral hemorrhage.3.The expression of MMP-9 and C3 in the area around hematoma was increased with the ICH.4.Xingnaojing Injection may protect blood-brain barrier and alleviate cerebral edema by decreasing the expression of MMP-9 and C3,and finally improve the neural function. |
