Objective:To observe the effect of different concentrations of BDE-209 on the human ovarian cancer SKOV3 cell,and its possible mechanism.Methods:The human ovarian SKOV3 cell was exposed by BDE-209 with the concentrations of 5ug/ml,10ug/ml,20ug/ml,then to be observed:(1) the effect of different concentrations of BDE-209 on the cell livability within 72hours by the MTT colorimetric analysis;(2) the cell morphologic changes respectively under the optical microscope and fluorescent microscope after the coloration of AO/EB;(3) the rate of early apotosis by Annexin V/PI of flow cytometry;(4) determination of a xanthine oxidase activity of SOD and determination of thiobarbituric acid content of MDA.Result:(1) The result of MTT colorimetric analysis show that exposure of BDE-209 can restrain the growth of the human ovarian cancer SKOV3 cell:only when the concentration of BDE-209≥10 ug/ml and exposed time≥48 hours,can it restrain the cell growth.We can see it obviously on the concentration of 20ug/ml,the cell livability of BDE-209 on 20ug/ml have significant difference from control group,P<0.05. (2)The exposure of BDE-209 have an effect on the cell morphologic changes:under the optical microscope,the shape of cells gradually change to round and the gap increased, the brightness of cell membrane reduced;cytoplasm have some granular and vacuole-like things;the cells showed collapse of status,particularly,morphological changes observed after 72h more apparent.Under fluorescent microscope:structure of the early apoptotic cell membrane is intact,nuclear is stained yellow green;the chromatin condensed and margined;the late apoptotic cell chromatin have orange-red flu- orescence and show margined or beaded-like,sometimes it is scattered in the cytoplasm .(3) The result of apoptosis rate was as follows by Annexin V/PI double staining test: at the same time,the rates of early apoptotic and necrosis cells in different experimental groups and the control group have significant difference,P<0.05;the experimental groups of the same concentration at different times compared to have significant differrence, P<0.05.As the time prolong,the concentration increased,the rates of early apoptosis and necrosis increased.(4)The exposure of BDE-209 on human ovarian cancer SKOV3 cells can induce intracellular SOD vitality reduced,cell MDA content increased.At the same time,the experimental groups compared with the control group there are significant differences,P<0.05;the experimental groups of same concentration at different times are statistically compared difference,P<0.05.As the concentration increased,the time prolong,cell SOD viability decreased;MDA content increaseed gradually,the cell oxidative stress increased,then can cause apoptosis and necrosis.Conclusion:The exposure of BDE-209 on human ovarian cancer SKOV3 cells have inhibitory effects,as the concentration increased,the time prolong,the apoptotic and necrosis rates increased,cell viability of SOD reduced and MDA content increased, the damnification of cells aggravated.
