| Objective:To investigate effects of naringin on expression of COX-2mRNA and protein ofHuman ovarian cancer cell line SKOV3.Methods:1ã€SKOV3cells were cultured in vitro and divided into five groups randomly,namely control group, naringin group(10μmol/L),naringin group(20μmol/L), naringingroup (40μmol/L) and celecoxib group(80μmol/L).2ã€The inhibition effects of naringin on cell-proliferation were determined byMTT method.3ã€The expression of COX-2mRNA were evaluated by real time quantitativePCR after Forty-eight hours.4ã€The expression of COX-2protein were evaluated by Western blot afterForty-eight hours.Results:1ã€Naringin could inhibit the growth of SKOV3cells, and with the increase ofthe dosage and time,the tumor growth inhibition rate increased significantly.The24hgrowth inhibition rates of naringin groups (10umol/L,20umol/L,40umol/L) were35.78%,59.63%,76.15%,and the48h growth inhibition rates were45.36%,67.12%,85.50%,and the72h growth inhibition rates were47.09%,75.87%,90.32%.Respectively,each was significantly higher than that of the control group(P<0.05).2ã€Compared with control group(1.094±0.053),the expression of COX-2mRNAin naringin and celecoxib groups were0.828±0.006,0.753±0.011,0.412±0.216,0.321±0.017,which were significantly lower than that of the control group(P<0.05),especially the40umol/L group and celecoxib group(P<0.01).3ã€Compared with control group(1.7325±0.0826),the expression of COX-2protein was higher by10μmol/L naringin group(1.7925±0.0880),but there was nostatistical difference. However, when SKOV3cells were treated by other dosage ofnaringin and celecoxib, the expression of COX-2protein were down-regulated significantly (1.2225±0.0822,1.2725±0.07632,0.3075±0.0977ï¼›P<0.05).Conclusions:The appropriate dosage of naringin can inhibit SKOV3cells growth in vitro, andcan down-regulate the expression of COX-2mRNA and protein. |
