Objecttive:To investigate the changes of plasma quantity of Tissue factor(TF) and Tissue factor pathway inhibitor(TFPI) in different type of Coronary heart disease(CHD) patients and the association between Tissue factor pathway and Coronary heart disease and the application prospects of using TFPI to cure the CHD patients.Methords:This study was conducted with 66 CHD patients and 22 healthy adults.The blood specimens were collected before being treated by Low molecular weight heparin(LMWH) in Acute coronary syndrome(ACS) patients,and the blood specimens of Stable angina pectoris(SAP) patients and healthy adults were collected on the second morning with a fasting status.After centrifugal treatment,the plasma was saved in a Ultra-low temperature refrigerator.Coronary angiography was carried out on each one of the selected objects,and the quantity levels of total cholesterol(TC),triglyceride(TG),high density lipoprotein -cholesterol(HDL-C) and low density lipoprotein -cholesterol(LDL-C) were detected.The plasma TF and TFPI quantity were measured by Enzyme-linked immunosorbent assay(ELISA).Results:The plasma TF antigen levels were highter in the Acute myocardial infarction(AMI) and Unstable angina pectoris(UAP) groups than in Stable angina pectoris(SAP) and control groups(161.08±20.90 and 152.76±20.66 pg/ml vs 99.72±16.75 and 94.32±12.93 pg/ml,p<0.05),there was no significant difference between the AMI group and UAP group,SAP group and control group(P>0.05).The plasma TFPI-1 antigen levels were bighter in the AM1 and UAP groups than in the SAP and control groups(32.05±8.52 and 31.49±10.61ng/ml vs 19.93±9.22 and 19.21±9.60 ng/ml,p<0.05),there was no significant difference between the AMI group and UAP group,SAP group and control group.The plasma TFPI-2 antigen levels were highter in the AMI and UAP groups than in the SAP and control groups(4.56±0.96 and 4.73±1.04 ng/ml vs 2.43±1.07 and 2.06±0.64 ng/ml,p<0.05), there was no significant difference between the AMI group and UAP group,SAP group and control group.The quantity of TF was higher in those patients who got a high score than those who got a lower score(121.15±27.38pg/ml VS 158.84±25.90pg/ml VS 187.10±8.10pg/ml,P<0.05 ).The quantities of plasma TF and TFPI-1 were higher in those patients who had 3 sick coronaries and 2 coronaries than those who had 1 sick coronary(P<0.05),there was no difference of TFPI-2 among the three groups(P>0.05).There was a positive relationship between the plasma TF and TFPI-1 and TFPI-2(r=0.755 and r=0.699,p<0.001).There existed a positive relationship between the quantity of plasma TF and the quantities of serum TC,LDL-C(r=0.563 and r=0.415,P<0.001),between the quantity of plasma TFPI-1 and the quantities of serum TC,LDL-C(r=0.633 and r=0.386,P<0.001),between the quantity of plasma TFPI-2 and the quantities of serum TC,LDL-C(r=0.248 and r=0.235,P<0.001),there were no significant relationship between TF,TFPI-1,TFPI-2 and TG,HDL-C(P>0.05).There was significant relationship between the plasma TF antigen level and the Gensini score of CHD patients(r=0.643,p<0.05),and there were no significant relationships between the plasma TFPI-1,TFPI-1 antigen level and the Gensini score(P>0.051.The plasma TF,TFPI-1,TFPI-2 antigen levels did not correlate with the extent of coronary atherosclerosis(P>0.05).Conclusions:The plasma TF,TFPI-1,TFPI-2 antigen levels of ACS patients are higher than those of SAP patients and healthy adults.This result indicates that ACS patients have an abnormally activity of coagulation system and the tissue factor pathway plays an important role in CHD patients.During our clinical work,we can reduce the incidence of coronarv event through actively controlling plasma levels of tissue factor pathway in CHD patients.There were positive relationships between plasma TF,TFPI-1,TFPI-2 quantities and serum TC,LDL-C quantities,indicates that we can reduce the activity of Tissue factor pathway through controlling the quantities of TC and LDL-C.There was a positive relationship between plasma TF level and the extent of coronary artery lesions,this indicates that plasma TF quantity may become another laboratory indicator which can evaluate the severity of CHD.
