Objective:Establishing C6 rat intracerebral glioma model,in order to investigate migration of the neural stem cells(NSCs) transplanted into the hippocampus of adult rat with intracerebral C6 gliomas.It will provide theoretical evidence for the experimentally therapy of gliomas.Methods:1.C6 brain malignant glioma cells were cultured in vitro.SD rat with intracranial C6 gliomas were established using stereotactic methods.2.24 rats were divided to a treatment group(n=12) and a control group(n=12) with random principle.The NSCs tagged with fluoresent Hoechst33342 were inplanted into the location of caudate nucleus in the treatment group,the number of the neural stem cells(NSCs) is 1.5×104 each rat.NS replace neural stem cells(NSCs), the rats were sacrificed at 7d,14d and 28d and the brains were examined by immunohistochemical analysis to observe the survival,migration of the NSCs in vivo.Results:1.C6 brain malignant glioma cells were inoculates with 20 days or so,the spirit of rat is appearing dejected,food and drink decreases too,about one month or so,many of the rats obviously hemiplegia,the spirit is greatly depressed,reacting is slow.Tumor was found in the brain of the rats.HE dying certify exist of tumor.2.It was obvious that the NSCs tagged with fluoresent Hoechst33342 at the location of hippocampus was observed in each times.neural stem cells(NSCs)could survive at the location of transplantion.Migration could not be obviously observed at 7d and 14d.The NSCs have obvious tropism for C6 glioma at 28d.the grafted NSCs may migrate along the lateral ventrical.Conclusion The C6 rat intracerebral glioma model is a stable and reliable brain glioma modle,the grafted NSCs and may tropism for glioma,but the primary mechanism neeed to study in future.
