ObjectiveIn this study,we discuss tropism and differentiation of implanted neural stem cells in vivo glioma,as with neural stem cells as a carrier therapy to provide a theoretical basis for glioma.Methods1.Cultivation of green fluorescent protein(GFP) neural stem cells and C6 glioma cells in vitro.2.SD rat with intracranial C6 gliomas were established using stereotactic methods.24 rats were divided to a experimental group(n=12) and a control group(n=12) with random principle.C6 glioma cells were planted into the location of under frontal cortex in the experimental group;cell culture medium were planted into the locatioh of under frontal cortex in the control group.Observe growth of the rats and determined by morphological detection of success or failure of rat tumor model.3.Tthe experimental group and control group SD rats are transplanted GFP neural stem cells in SVZ.Using inverted fluorescent microscope,we observe migration of transplanted neural stem cells in vivo.Inverted fluorescence microscope,flow cytometry analysis and observed exogenous neural stem cells in vivo survival, migration and differentiation.Results1.Isolation and culture of the GFP neural stem cells into cell clusters neurospheres grew,the growth of primary cells after 3~4d suitable passage.After third passage 4d obtained with a large number of positive cells expressed nestin antibody.2.Brain scan and MRI,immunohistochemistry,confirm the formation of solid tumors in the rat brain,a successful model.3.Observe under the fluorescence microscope,experimental group and the surrounding areas within the tumor tissue visible GFP. neural stem cells.4.By flow cytometry examination:In the tumor area in experimental rat models GFAP(+)/GFP(+)cells are more than that in transplant area(P<0.05),and this trend becomestronger when the time goes on(P<0.05).Conclusion1.The NSCs isolated and cultured by using the present methods have the ability of self-renewal,and flow cytometry examination:expression of nestin antibody were positive.So Proved to be the central nervous system stem cells.2.The C6 rat intracerebral glioma model is stable and reliable brain glioma model. rat C6 glioma tumor growth in animal models has certain ragulation,their growth characteristics and pathological features similar to human gliomas,gliomas can be used as basic and clinical research of the ideal model.3.In glioma model,by morphological and flow cytometry analysis the transplanted neural stem cells migrate towards the tumor area.4.Flow cytometry results show,stem cell transplantation relative to the region, migration to the tumor periphery of neural stem cells clear trend in the glial cells(P<0.05),and within the range of 7~14 days,as time advances,the more strengthen to the glial cells(P<0.05).
