Objective:To explore whether Nomifensine has the neuroprotective effects on substantianigra striatum's dopaminergic neurons in subacute MA poisoning rat and its mechanism.Methods:72 Wistar male rats weighing were randomly divided into saline group,MA group and MA+Nomifensine group(protective drug group).Saline group was treated with saline 10mg/kg intraperitoneal injection at 8:00 pm each day.MA group was treated with MA 10mg/kg intraperitoneal injection at 8:00 pm each day.MA+Nomifensine group was treated with both MA 10mg/kg and Nomifensine 2mg/kg intraperitoneal injection at 8:00pm each day.Administration time was 8:00pm everyday.Four days to build subacute MA poisoning model,we used general state of health,stereotyped behavior and Conditioned Place Preference(CPP) to evaluate whether the model was successful.We killed the successful model rats at the first,seventh,fourteenth days after the last administration,using both immunohistochemical method and fluorescence spectrophotometer which can detect their dopaminergic neurons' form and quantity of substantia nigra,midbrain ventral tegmental area,cortex of frontal lobe,hippocampus,striatum,nucleus accumbens septum in morphological, semiquantitative and quantitative aspects.We used image analysis system to calculate masculine cells and analyze gray scale value,in addition we compared the date of different groups with statistical treatment.Results:1.The research has shown obvious statistical significance between negative control group and positive control group,protective drug group in stereotyped behavior scores and duration(P<0.01).The research has also shown obvious statistical significance between protective drug group and positive control group in stereotyped behavior scores and duration(P<0.05andP<0.01) too. However they have shown obvious statistical significance between protective drug group and positive control group in CPP results(P<0.05).2.At different brain regions and different time points by counting neurons,nerve fibers of gray value and TH content fluorescence spectrophotometer measurement results have shown that drug group to protect the nervous system damage was lighter than positive control group.Conclusion:Nomifensine shows some neuroprotective effect in Subacute MA poisoning rat.
