More and more research have shown that MPTP plays a important role in the etiology of PD.While the latest studies have identified that the MPTP is oxidased to MPP~ by Monoamine oxidase B (MAO-B) which is a kind of enzyme in membrane. MPP is what really works.Many in vivo studies have proved MPTP can cause PD.We are interested in investigating the influence of MPP+ on mesencephalic Dopamine neurons in vitro and to make sure whether it can be used as an PD model in vitro.In order to explore the function of glia in the progress the experiment was performed both in presence of glia and in the absence of glia The mouse mesencephalic cells were exposed with MPP+ with different concentration. Iloprost was used to investigate whether it can prevent dopamine neuron from the neurotoxic of MPP~. .Then they were tested for [3H] DA and PH] Choline uptake and intracellular DA content ,as well as the TH immunireactivity. The result are as follow: 1. The treatment of mouse embryo mensencephalic cell cultures with MPP~ reduced the PH] DA uptake no matter in the presence or in the absence of glia but not to the PH] Choline uptake.The neurotoxic effect was more pronounced in the presence of glia as compared with in the absence of glia. 2. Not only the DA content but also the number of TH positive cells significantly decreased in the mouse embryo mensencephalic cells when being treated with MPP+. 3. The were no difference between different concertration of MPP~. 4. Iloprost failed to protect dopamine neuron from the neurotoxic of MPP+. From the result above we believe :The treatment of mouse embryo mensencephalic cell cutures with MIPP+ reduced the viability of dopaniinergic neurons but not to the acetylcholinergic neurons .MPP+may be used to produce PD model in vitro.
