Establishment And Analysis Of The Finger Print Chromatogram For Liver-regulating Compound Recipe JWSNS

Objective:In the present study,the research aim is to establish the finger print chromatogram of liver-regulating compound recipe called Jiaweisinisan(JWSNS).Methods:With the method of Standard decoction,we got the sample for the high performance liquid chromatography(HPLC).We analyze ten batch samples with HPLC to establish the JWSNS HPLC finger print chromatogram.The method of Standard decoction is like below.First,we weigh abalone for thirty gram,and add them into water which about 600ml,soak them about 30 minute,and then boil them first,2100w 3min,800w 30min.At this time soak the other six part of JWSNS for 30min.After the first boiling we measure the remaining physic liquor A.Interpolate some water(I.e 670ml-400ml-A) into the holder,after that we begin the second turn boiling,which the way is same with the first time.After the second boiling we measure the remaining physic liquor B.And then add 520ml water into the holder,2100w 3min,800w 25min.After the third boiling we also measure the remaining physic liquor C.At last,we get together with A and B.And then we affiliate the physic liquor from three holder,and filter them when they are hot,get the one third physic liquor as sample.The pattern number of chromato bar is Phenomenex LunaC₁₈(2)(250×4.6mm,5μm) with diode array detector.Acetonitrile-0.1%orthophosphoric acid as mobile phase,radient elution,rout flow rate is 1.0 ml/min,wavelength is 230nm, column temperature is 30℃.With the State Pharmacopeia Commission traditional Chinese drug finger print chromatogram semblance appreciation system software(2004A),we analyzed ten batch samples and got the mutual peak,and identify the composition of characteristic absorption band by ultraviolet spectra and reference substance.Results:The relative standard deviation(RSD) of twenty-two batches Standard decoction is 8 percent.The RSD of ten batch is 8%,and of the seven single part is 9%,the five repeat one have 3%RSD.The semblance of ten batch samples is beyond 0.98.The result of peak position adscription displays the resource of JWSNS finger print chromatogram.Eight ones belongs to White peony Alba,nine ones belongs to cape jasmine fruit,fourteen ones belongs to Orange Fruit,one from rehmannia dride rhizome,one from Lycium chinense Mill.There are four peak were identified,the tenth peak is jasminoidin,the fourteenth one is peoniflorin,the eighteenth one is naringin,the nineth one is aurantiamarin.Conclusion:the relative standard deviation of 8 percent of Standard decoction is satisfied with precision requirement of HPLC finger print chromatogram(RSD＜3%Chinese crude drug injection finger print chromatogram specification).The characteristic absorption band of JWSNS HPLC finger print chromatogram is synthetics of singal composition,there is not a new conspicuous one.The most of them comes from White peony Alba,jasmine fruit, Orange Fruit.
