High performance liquid chromatography has the advantage of high pressure,high efficiency,high sensitivity and simple operation.It has wid e application in the modern analysis field,such as Chinese medicine,food sanitation,biological engineering,environmental safety,petrochemical industry and so on.The principle of high performance liquid chromatography is the adsorption and unadsorption velocity difference of analytes in the chromatographic column.In practice,the analyte must undergo a rigorous pretreatment process before high performance liquid chromatography(HPLC),for achieving the purpose of effective extraction and enrichment of trace components and achieving more accurate experimental data,a variety of sample pretreatment methods emerge as the times require.Environment friendly,simple,fast sample preparation technology for determination analysis is always the target that the plenty of researchers pursue.The paper mainly includes two parts: review and research report.In the first part,the development and application of high performance liquid chromatography,Chinese herbal medicine,flavones,organic acid and sample preparation was reviewed.In the research report part,the determination method of f lavonoids compounds and organic acid in diffrent Chinese traditional medicine was established.1.Determination of Six Flavonoids in Agrimonia Pilosa Ledeb by HPLCThe HPLC method was established for separation and determination of rutin,taxifolin,coumarin,luteolin,kaempferol and apigenin in Agrimonia Pilosa Ledeb by ultrasonic extraction.The separation of six flavonoids was performed on Phenomenex C18 column(150 mm×4.6 mm,5 ?m)with step gradient.Mobile phase was methanol-0.1% acetic acid at the flow rate was 0.8 m L/min;the UV detection wavelength was 320 nm and the coclumn temperature was set at 35 ?. The good separation of rutin,taxifolin,coumarin,luteolin,apigenin and kaempferol was achieved within 15 min.Calibration curves of the six components showed linear relationship(r>0.999 1,n=6).The average recoveries were within 97.78%-104.4%(RSD<1.20%).This ultrasonic extraction method was simple,fast,economical and reliable.2.Determination of Six Flavonoids in Agrimonia Pilosa Ledeb by HPLCThe HPLC method was established for separation and determination of epicatechin,gallic acid,catechin,rutin,myricetin,quercetin and kaempferol in Toona sinensis by ultrasonic extraction.The separation of seven active ingredients was performed on Phenomenex C18 column(150 mm×4.6 mm,5 ?m)with step gradient.Mobile phase was methanol-0.1% acetic acid at the flow rate was 0.8 m L?min-1;the UV detection wavelength was 290 nm and the coclumn temperature was set at 35?.The good separation of epicatechin,gallic acid,catechin,rutin,myricetin,quercetin and kaempferol was achieved within 15 min.Calibration curves of the seven active ingrediens showed linear relationship(r>0.9991,n=7).The average recoveries were within 99.23% ~ 104.5%(RSD%<1.26%,n=3).This ultrasonic extraction method was simple,fast,economical and reliable.3.Determination of Six active components in Viola yedoensis Makino by HPLCThe HPLC method was established for separation and determination of cichorigenin,caffeic acid,rutin,naringenin,luteolin and kaempferol in Viola yedoensis Makino by ultrasonic extraction.The separation of six active components was performed on Phenomenex C18 column(150 mm×4.6 mm,5 ?m)with step gradient.Mobile phase was methanol-0.1% acetic acid at the flow rate was 0.8 m L?min-1;the UV detection wavelength was 340 nm and the coclumn temperature was set at 35?.The good separation of cichorigenin,caffeic acid,rutin,naringenin,luteolin and kaempferol was achieved within 15 min.Calibration curves of the six components showed linear relationship(r>0.9991,n=8).The average recoveries were within 102.13%~104.55%(RSD%<1.20%,n=3).This ultrasonic extraction method was simple,fast,economical and reliable.4.Determination of 6 Active Ingredienst in 6Species of Chinese Medicinal Herbs by Inonic Liquid-based Ultrasoninc-assiststed Combined with High Performannce LiquidThe HPLC method was established for separation and determinati on of Chlorogenic acid,utin,quercetin,Luteolin,kaempferol and Apigenin in Artemisia argyi Levl.et Vant,Plantago depressa Willd,herba houttuyniae,Nepeta cataria L.,Scutellaria barbata D.Don,Taraxacum mongolicum Hand.-Mazz.by ionic liquid-based ultrasonic extraction.The separation of six active ingredients was performed on Phenomenex C18 column(150 mm×4.6 mm,5 ?m)with step gradient.Mobile phase was methanol-0.1% acetic acid at the flow rate was 0.8 m L?min-1;the UV detection wavelength was 290 nm and the coclumn temperature was set at 35?.The good separation of Chlorogenic acid,utin,quercetin,Luteolin,kaempferol was achieved within 13 min.Calibration curves of the seven active ingrediens showed linear relationship(r>0.9991,n=7).The average recoveries were within 99.68% ~ 104.9%(RSD%<1.70%,n=3).This ultrasonic extraction method was simple,fast,economical and reliable.
