Objective To study the proliferation ability of CIK(Cytokine-induced kill cells, CIK) cells in vitro, and to investigate the anti-tumor effects of CIK,paclitaxel and CIK combined with paclitaxel against gastric cancer cell lines SGC-7901 in vitro.Methods The peripheral blood mononuclear cells (PBMC) of healthy human were induced with different cytokines, and transfered in CIK cells. The proliferation ability of CIK cells was measured by calculation in different culturing time, and the phenotype of CIK cells was analyzed by a flow cytometer. Inverted microscope was used to observe morphological changes of SGC-7901 gastric cancer cell after added the CIK cells. The CIK cell culture supernatant and paclitaxel induced gastric cancer cells apoptosis was analyzed by a flow cytometer. The antitumor cytotoxicity of CIK cells and paclitaxel against SGC-7901 cell lines was measured by MTT assay, and the anti-tumor effects of CIK cells combined with paclitaxel were investigated.Results With the cultured time of CIK cells prolong, both the number and killing activity increased;CIK cells possessed strong proliferate ability with about 111.63±10.97 flod expansion after 21 days culture, CD3+CD56+ double positive cells in the ratio was up to 35.8±9.7%, and then, both the proliferate ability of CIK cells and the percentage of CD3+CD56+ cells declined. CIK cells possessed significant ability to kill SGC-7901 tumor cells; the maximum destruction efficiency is about 74.91±2.71%. Both the CIK cell supernatant and paclitaxel can induce apoptosis of gastric cancer cell. When CIK combined with paclitaxel, the killing effect was enhanced.Conclusions CIK cells own strong anti-cancer activity against SGC-7901 gastric cancer cells in vitro. The suitable time for clinical application is at day 14 to 21, which is the proliferation peak. When CIK cells combined with Taxol, they showed synergistic killing effects.
