Objective Discussion targeting NF-κB oligonucleotide(ODN) "trap" strategy of acute pancreatitis rat pancreas damage the protective effect and mechanism.Methods Wistar rats with 36,were randomly divided into 3 groups:respectively,the former model in the tail vein injection of NF-κB oligonucleotide targeting group,injection targeting NF-κB oligonucleotide missense chain group and no injection any drug control group.Animals in each group after modeling 6h,12h,24h were killed in three batches,take pancreatic tissue and serum,light microscope to observe the extent of pancreatic tissue injury changes,the use of ELISA method of expression of serum TNF,immunohistochemical method NF-κB activity. Trial period were observed in rats and mortality in the general situation.ResultsⅰThree groups of rats were successfully model the experimental period,survival, mortality 0%,the spirit of survival period,activities were poor,B group,C group than in group A re.ⅱAnimals in each group after modeling 6h,12h,24h were killed in three batches,take pancreatic tissue and serum,light microscope to observe the extent of pancreatic tissue injury changes,the use of ELISA method of expression of serum TNF,immunohistochemical method NF-κB activity.A group of pancreatic cells appear under a light microscope edema,degeneration and necrosis,but more B group,C group the degree of light,ⅲAt the same time with A group, B group,C group the average level of TNF in general there is a difference,compared to 22,A group and B group are a few differences were statistically significant(<0.001),that the B group with high levels of TNF in the A group;A group and C the number of inter-group differences were statistically significant(<0.001),that the C group than the A group TNF levels;B group and C group could not believe that there are significant differences,ⅳA group of post-model, B group,C group pancreatic tissue expression of NF-κB-positive rate,positive rate of three groups were statistically significant differences,compared to 22,A group and B group were statistically significant differences(=0.013),that the positive rate of group B higher than the A group;A group and C group were statistically significant differences(=0.003),that the positive rate of C group than the A group;B group and C inter-group differences can not believe that there are statistically significant(=0.537).Conclusion Targeting NF-κB oligonucleotide "decoy" strategy can bc specifically inhibited the activity of NF-κB,reduced the level of TNF,pancreatitis rats with acute pancreatitis of injury has a protective effect.
