The Study On The Antitumor Activity Of The Immunosuppressive Agent FTY720 To The Rat Hepatocellular Transplanted Tumor

Objective To study the antitumor activity of the immunosuppressive agent FTY720 to the rat hepatocellular transplanted tumor.Methods Hepa1 - 6 cells were collected and injected subcutaneously with a dose of 2×106 /mouse into C57BL /6 jmice.When the diameter of the hepatocellular transplanted tumor achieving to 0.30cm,the 40 C57BL /6 j mice were randomly assigned Into 4 groups, Each group was lavaged with the medicine separately such as (1)ciclosporin 10 mg·kg-1·d-1 (2)FTY720 3mg·kg-1·d-1;(3)5-fu 50 mg·kg-1·d-1; (4)0.9% NS 50 ml·kg-1·d-1.After 10 days,thymus gland and spleen index ,the percentage of T-lymphocyte ,gross tumor volume, secretory volume of AFP in serum,apoptosis index ,the expression of Caspase-3 and Bcl-2 were investigated.Results thymus gland and spleen index and the percentage of T-1ymphocyte in mice were Iower in the ciclosporin group and FTY720 group than the saline group (P <0.05,P <0.01),There were no significant diferences between saline group and 5-fu group(P>0.05);The gross tumor volume and secretory volume of AFP in serum were Iower in the 5-fu group and FTY720 group than the saline group (P <0.05,P <0.01)and the apoptosis index is higher 5-fu group and FTY720 group than the saline group (P <0.05,P <0.01), There were no significant diferences between saline group and ciclosporin group(P>0.05);The expression of Bcl-2 is Iower in the 5-fu group and FTY720 group than the saline group(P<0.05), there were no significant diferences between saline group and ciclosporin group(P>0.05); The expression of Caspase-3 is Iower in the 5-fu group than the saline group(P<0.01), There were no significant diferences between saline group and FTY720 group,ciclosporin group(P>0.05).Conclusion FTY720 have immunosuppression and antitumor activity,the antitumor activity is connected with the lower expression of Bcl-2.
