Objective FTY720, a synthetic compound produced by modification of a metabolite from Isaria Sinclair, is known as a unique immunosuppressive agent that were widely used in clinical organ transplantation.Additionally, it has been found that FTY720 has inhibitory effect on various cancer growth and metastasis. To investigate its effect on the growth and metastasis of hepatocellular carcinoma Hepal-6, and explore its mechanism.Methods The hepatocellular carcinoma Hepal-6 were treated with FTY720 at various concentrations (0~100μg/ml)in present study. The MTT assay was used to assess cytostatic activity induced by FTY720. Flow cytometry(FCM) was adopted to evaluate the cell apoptosis after FTY720 treament. Immunohistochemistry (IHC)was used to demonstrate expression levels of involved such as Bcl-2 and caspase-3.Results The MTT assay indicated that the growth of hepatocellular carcinoma Hepal-6 could be inhibited by FTY720 at various concentrations between 0~100.0μg/ml in a does-depentdent manner, which was also confirmed by FCM. After FTY720 treatment, the Bcl-2 were down regulated while the caspase-3 were not significant change.Conclusions In this study, the apoptosis-induced effects of FTY720 on hepatocellular carcinoma Hepal-6 were analyzed. The FTY720 can inhibit growth, also can induce apoptosis of mice hepatocellular carcinoma. The finding presented here revel that FTY720 induce marked apoptosis in mice hepatocellular carcinoma mainly by influencing Bcl-2.
