Objective: To research the effect of urokinase-type plasminogen activator receptor (uPAR) on human sperm viability, motility rate, and sperm acrosome reaction(AR) and investigate the mechanism.Methods: Sperm were aseptically obtained by masturbation and prepared by swim-up technique to produce a spermatozoa solution of high motility from 20 healthy fertile men. Part one the spermatozoa solution was incubated with the different concentrations of anti-uPAR antibody(10μg/ml,50μg/ml,100μg/ml) in vitro. Measurements were carried out at 10, 30 and 60 min in 10 specimens, including sperm motility, sperm viability and motility function, while establishing normal control group. Part two, the spermatozoa solution which were treated by swim-up technique were incubated with the different concentrations of. anti-uPAR antibody(10μg/ml,50μg/ml,100μg/ml) at 37℃in vitro. The FITC-PSA was used to stain the capacitated spermatozoa that have be incubated for 60 min in vitro. Acrosome reaction induced by anti-uPAR antibody was measured by means of a flowcytometer, while establishing normal control group.Results:1), Part one, After 10min,each group,s viability and motility rate have no significantly different (P>0.05);compared with the control group, 10μg/ml group's viability and motility rate have no significantly different too(P>0.05). But after 30min and 60min, 50μg/ml group and 100μg/ml group's viability and motility rate have significantly different, compared with the control group(P<0.05).2), Part two, compared with the control group, 10μg/ml group, 50μg/ml group, 100μg/ml group, the fluorescent marker values were higher. The fluorescent marker values have significantly different(P﹤0.01). There were not significantly different(P﹥0.05) between 10μg/ml group , 50μg/ml group and 100μg/ml group.Conclusions:1,Anti-uPAR antibody of different concentrations can reduce human sperm viability, motility rate and motility function. 2,Anti-uPAR antibody of high concentration can inhibit acrosome reaction.3,uPA/uPAR can effect the sperm viability, motility rate, and sperm acrosome reaction(AR) and promote intracellular calcium influx in vitro.4,after capacitation calcitonin would begin to inhibit spontaneous acrosome reactions.The responses obtained in vitro suggest that this peptide could also be used effectively in infertility research and clinic.
