Backgroud Anemia of chronic disease is a common anemia clinically,the incidence of anemia of chronic disease is second only to the incidence of the iron deficiency anemia.The anemia affects obviously the quality of life about anemia patient. In the past,it is an orthodox way to improve the anemia by blood transfusion. But mang questions curse patient,they are high expense poor effect,lack of blood,infection,transfusion reaction,immune suppression and so on. People are concerned about the questions,how to improve the quality of life of the patients and prolong survival time of patients. Therefore the pathogenesis of anemia of chronic disease has become the focus of the study. Recently people think that IL-6 and hepcidin may play a role in the pathogenesis of anemia of chronic disease,this may provide a new way to cure anemia. Objective On the basement of establishing a rat model of anemia of chronic disease(ACD),investigating the IL-6 and hepcidin of the pathogenesis in the ACD to provide a theoretical basis for prevention and treatment.Methods 1. According to the interventions, twenty-four rats were randomly divided into three groups,every group number was eight, called normal control group(N group),ACD model group(M group),EPO therapy group(L group).The taditional animal model of rheumatoid arthritic was established by injecting Freund's complete adjuvant into rats only time .On the basis of the model, we injected Freund's complete adjuvant thirds again so that anemia was induced, Detection of blood, pigmented cells are being anemia in the 60th days. That the rat model of was established. 2.The different groups were treated with EPO or NS; injected 2000IU/kg every for three days.Execute the rats after 24 hours. 3. Observe the swelling extent change of the toe joints and their general state of rats,Acquisition of fresh blood and liver tissue,detection blood,liver hepcidin mRNA expression and blood IL-6 level.Results In the ACD model group IL-6 in the serum of rats and the expression of hepcidin mRNA in the livers of rats was more than in the normal control group(p<0.05), anemia is significant,The relativity of serum IL-6 and Hb in the ACD model group is negative (the coefficient is 0.93),The relativity of serum hepcidin mRNA and Hb in the ACD model group is negative(the coefficient is 0.958),While in the EPO therapy group the expression of hepcidin mRNA and IL-6 in the serum of rats was less than in the ACD model group(p<0.05),anemia is improved.In the EPO therapy group the expression of hepcidin mRNA in the livers of rats was less than in the normal control group(p<0.05).The relativity of serum hepcidin mRNA and IL-6 is positive(the coefficient is 0.979). Conclusions 1. Anemia of chronic disease rats model was induced by injecting the Freund's complete adjuvant into adjuvant rats. 2. Hepcidin and interleukin -6 may play an important role in pathogenesis of ACD . 3. It is a good way to prevent the development of anemia to decrease hepcidin and interleukin-6 level, It will provide a new way for the treatment of ACD. 4. Erythropoietin may improve anemia by inhibiting the IL-6 to decrease the hepcidin.
