Objective:To compare the LOH rate of allelomorphic gene on chromosome 9 at the identical district and site between esophageal squamous carcinoma and cervical squamous carcinoma to explore the related anti-oncogene in common.Methods:LOH of seven microsateUite markers on chromosome 9 were detected in 45 cases of esophageal squamous carcinoma and 46 cases of cervical squamous carcinoma by microdissection,PCR,denaturing polyacrylamide gel electrophoresis,silver nitrate staining and radiograph technology.The relation of LOH rate and clinical pathology parameter were analyzed using x~2 test by SPSS13.0 software.Results:①In the informative cases,the total frequency of LOH in esophageal squamous carcinoma was 25.8%,LOH was detected as follows:43%at D9S753,38%at D9S303,36%at D9S171,36%at D9S162,17%at D9S242,17%at D9S1748 and 0%at D9S43.The total frequency of LOH in esophageal high-grade intraepithelial neoplasia was 16.4%,LOH was detected at marker 30.8%at D9S303,28.6%at D9S171,28.6%at D9S753,15.1%at D9S162, 11.1%at D9S242,6.7%at D9S1748and 0%at D9S43.②In the informative cases,the total frequency of LOH in cervical squamous carcinoma was 25.2%,LOH was detected as follows: 40.7%at D9S171,33.3%at D9S43,31.4%at D9S162,23.7%at D9S242,17.2%at D9S303,16.7%at D9S753 and 12.9%at D9S1748.The total frequency of LOH in cervical high-grade intraepithelial neoplasia was 15.9%,LOH was detected at marker 29.6%at D9S171,22.9%at D9S162,20.0%at D9S43,17.2%at D9S303,12.5%at D9S753,10.5%at D9S242 and 0%at D9S1748.③There was no significant association between LOH rate at seven microsatellite markers and gender,histopathologic grade,the information of lymphaden metastasis.Conclusions:①The study in seven microsatellite markers indicates that there is accumulation of anti-oncogene abnormality during the process from normal squamous epithelium to high-grade intraepithelial neoplasia and esophageal squamous carcinoma and cervical squamous carcinoma.②The abnormality of p15 is possibly associated with the development and progression of esophageal squamous carcinoma and cervical squamous carcinoma.③There are common anti-oncogene possibly at or near D9S162 which related to the progression of esophageal squamous carcinoma and cervical squamou.s carcinoma.④D9S303 and D9S753 on 9q13-22.3 possibly relate to progression of esophageal squamous carcinoma only.⑤D9S242 on 9q32 and D9S43 on 9p21-qter possibly relate to progression of cervical squamous carcinoma only.
