Background and Aim:It is conventional to use the combination of two classification systems including"Cervical intraepithelial neoplasia "and“High/low-grade squamous epithelial lesion"to diagnose the precancerous lesions of the cervix,which can be specifically classified into:LSIL/CIN1,HSIL/CIN2 and HSIL/CIN3.However,this new combined system still lacks objective criteria for the diagnose of CIN due to it continues to take the extent of the lesion accumulation as the only diagnostic basis.Therefore,there are quite a number of patients whose result of colposcopy biopsy is ambiguous between CIN2 to CIN3 even if which biological behaviors are distinctly different.It will inevitably lead to much excessive treatment if we manage above all patients of CIN2-3 according to CIN3.So it is crucial for us to search for an appropriate method to distinguish the diagnose of HSIL/CIN2-3 in a clear grade.At present,a large number of studies have been shown that SOX2 is closely related to the occurrence,development,differentiation,metastasis and poor prognosis of malignant tumors,and its expression can increase with the progression of malignant cells.The aim of this research is:1)to analyze the correlation between age,menopause and different cervical conization methods and postoperative pathology;2)to study the correlation between the expression of SOX2 in biopsy tissue and postoperative pathology.Materials and Methods:We chose the patients who have had colposcopy cervical biopsy with pathological result being HSIL/CIN2-3 and received cervical conization treatment at the Women's Hospital of Zhejiang University from January 1,2013 to December 31,2014 in further.Then,we retrospectively reviewed the age,menopause and different cervical conization approaches of patients and analyzed the relationship between postoperative pathology and above factors.At last,we recut off the paraffin-embedded tissue blocks of the included patients and processed them with immunohistochemical staining of SOX2.All data we got were statistical analyzed by SPSS 20.0.Results:The SOX2 immunohistochemical staining results in the patients with whose diagnose of colposcopy cervical biopsy tissue is HSIL/CIN2-3 were correlated significantly with postoperative pathology by univariate analysis and multivariate analysis(p=0.01<0.05).The age,menopause and different cervical conization approaches were not correlated with postoperative pathology(respectively,p=0.31,0.41,0.09).Conclusion:The SOX2 immunohistochemical staining results in the patients with whose diagnose of colposcopy cervical biopsy tissue is HSIL/CIN2-3 were correlated significantly with postoperative pathology.Presumably,SOX2 might be a promising candidate for the stratification of high-grade cervical intraepithelial neoplasia 2-3.
