ObjectiveScreening of differential expression genes between tumors and normal tissues by microarray is the hot of cancer research nowadays.VEGF-C,PDGF-A,SPARC are the differential expression genes of OSCC which have been selected by microarray.The study is to investigate the expression of the three differential expression genes in oral squamous cell carcinoma(OSCC) and their relationship to clinicopathological factors,to investigate their correlation,and to provide the new targets for the prevention and treatment of OSCC.MethodsImmunohistochemistry method(universal two-step method) was employed to determine the expression of VEGF-C,PDGF-A and SPARC in 60 cases of OSCC and epithelium immediately adjacent to carcinoma(EAC) respectively.Then the results were evaluated by computer aided image analysis system and the statistical analyses of average optical density(AOD) were done.The relationship between the three factors and the clinicopathological factors,such as gender,age,tumor site,T classification, pathology grading,regional lymphnodes and clinical stage,was evaluated by Spearman correlation analysis.Results1.The expressions of VEGF-C in OSCC were significantly higher than in EAC(u=7.747,P<0.01).The expression of VEGF-C was positively related with clinical stage(r=0.564,P<0.05) and regional lymph node metastasis(r=0.706,P<0.01),but no distinct difference with gender,age,tumor site,T classification,and pathology grading in 60 cases of OSCC.2.The expressions of PDGF-A in OSCC were significantly higher than that in EAC(u=6.982,P<0.01).The expression of PDGF-A was positively related with the differentiation of pathological grading(r=0.286,P < 0.05),regional lymph node metastasis,(r=0.314,P<0.05) but no distinct difference with gender,age,T classification,tumor site and clinical stage in OSCC.3.The expressions of SPARC in OSCC were significantly higher than in EAC(u=8.427,P<0.01).The expression of SPARC was positively related with clinical stage(r=0.375,P<0.01) and regional lymph node metastasis(r=0.495,P<0.01),but no distinct difference with gender,age,T classification,tumor site,and pathology grading in OSCC.4.The expression of SPARC had obvious correlated with VEGF-C and PDGF-A (r=0.570,0.501,P<0.01).The expression of VEGF-C had no obvious correlated with PDGF-A(r=0.172,P>0.05).Conclusion1.The expressions of VEGF-C,PDGF-A and SPARC were significantly higher,and were OSCC associated gene.2.VEGF-C,PDGF-A and SPARC are risk factors of lymph node metastasis in OSCC,examination of them can estimate the lymph node metastasis3.SPARC maybe has mutual concurrent effects with VEGF-C and PDGF-A,and the exact mechanism needs further study.4.It may become new targets for the gene therapy of OSCC by regulating the above indexes.
