Objective To investigate the effect of simvastatin on cardiac muscle and expression of myocardial cytokine in rats with cardiac hypertrophy.Methods The artial abdominal aorta ligature was used to induce hypertrophy in rats. Wistar rats were randomly divided into four groups:normal control rats, cardiac hypertrophic rats,cardiac hypertrophic rats treated wih simvastatin (5mg/kg.d),cardiac hypertrophic rats treated with lorsartan(2mg/kg.d), normal control rat and cardiac hypertrophic rats are treated wih normal sodium(9g/L)for eight weeks at the same time.Systolic blood pressure(SBP) was measured using the tail-cuff method.Myocardial cytokine expression was analyzed by real-time PCR,The heart weight,the ratio of heart weight to body weight were calculated,The pathobiological change of the hearts was measured.Results Compared with control group,Myocardial cytokine brain natriuretic peptide(BNP),interleukin-6/interleukin-10(IL-6/IL-10),the heart weight,the ratio of heart weight to body weight,the left ventricular wall thinkness and myocyte diameter are different in cardiac hypertrophy group (P<0.05 ).Compared with cardiac hypertrophy group,myocardial cytokine BNP,IL-6,IL-10 mRNA expression,the heart weight,the ratio of heart weight to body weight,the left ventricular wall thinkness and myocyte diameter are different in cardiac hypertrophic rats treated wih simvastatin or lorsartan(P<0.05).Conclusions simvastatin may regulate cardiac hypertrophy by inhibiting production of pro-inflammatory cytokine and raising expression of anti inflammatory cytokine in the heart.
