Objective:to observe the apoptosis of hepatoma carcinoma cell HepG2 which was induced by Xiaozhiling injection(XZL) in vitro,through analysis the expression of P53,Bcl-2 and nm-23 in HepG2,not only to preliminarily discuss the mechanism of action,but also to offer the feasibility of assessment and theoretical basis for treatment the primary hepatic carcinoma with XZL in clinical.Methods:to culture hepatoma carcinoma cells HepG2 in vitro with different concentrations of XZL(0.5,1,2,4,8,16g/l),and detect cell inhibition ratio by cytotoxicity test(MTT),then Western blotting was used to detect the change of correlated apoptosis protein P53,Bcl-2 and nm-23.then compared with the control group and positive control group.Results:(1) MTT showed that XZL could produce an obvious time-and-dose-dependent inhibitory effect on the HepG2 cells,The live cells decreased gradually with the increasing of drug concentration and disposal time.(2) The Western blotting confirmed that,compared with the control group,The expression of P53 protein increased,Bcl-2 protein decreased in the XZL group,the differences was significant(p<0.05).but there are no significant difference in the expression of nm23-H1 protein(P>0.05).compared with the 5-Fu group, the expression of Bcl-2 protein decrease,the differences was significant(p<0.05).but there are no significant difference in the expression of P53 protein and nm23-H1 protein(P>0.05). compared with the 5-Fu,XZL group,The expression of Bcl-2 protein increased,P53 protein decreased in the XZL group,the differences was significant(p<0.05),but there are no significant difference in the expression of nm23-H1 protein(P>0.05).Conclusion:XZLcould not only inhibit HepG2 cell multiplication,but also induce the apoptosis in HepG2 by increased the expression of P53 protein and decreased the expression of Bcl-2 protein.
