ObjectiveTo study the antiemetic effect and mechanism of gingerols.Methods1.After a pica model of rat was established with 1-phenylbiguanide hydrochloride (PBG),apomorphine and cisplatin,animals were randomly divided into normal control group,model group,positive control group and three treated groups(n = 6). The inhibitive effect of gingerols on kaolin pica behavior and consumption of rats was observed.2.After a vomiting model of mink was established with PBG and cisplatin,animals were randomly divided into normal control group,model group,positive control group and three treated groups(n = 6).The inhibitive effect of gingerols on vomiting behavior of minks was observed.3.Minks were killed 24 h after administration of cisplatin and the ileal 5-HT levels were measured by high performance liquid chromatography with electrochemical detection(HPLC-ECD).Results1.A pica model of rat was established with the specific 5-HT3-receptor agonist 1-phenylbiguanide hydrochloride 25 mg·kg-1.Gingerols exhibited inhibitive effect on kaolin pica behavior of rats and reduced kaolin pica consumption induced by apomorphine,PBG and cisplatin(P <0.05).2.A vomiting model of mink was established with the specific 5-HT3-receptor agonist PBG 8.0 mg·kg-1.Gingerols displayed inhibitive effect on vomiting behavior of minks induced by PBG and cisplatin in terms of prolonging the latency time of onset to vomiting and reducing the number of retches and vomits(P <0.05).3.The result of ileal HPLC in minks manifested that cisplatin caused a release of 5-HT from ileum and induced an increase in 5-HT levels(P <0.05).The increase in ileal 5-HT release induced by cisplatin was inhibited by gingerols(P <0.05).Conclusion1.The specific 5-HT3-receptor agonist PBG 25 mg·kg-1 induces a pica model of rat. Gingerols exhibit inhibitive effect on kaolin pica behavior of rats.2.The specific 5-HT3-receptor agonist PBG 8 mg·kg-1 induces a vomiting model of mink.Gingerols exhibit antiemetic effect on vomiting behavior of minks. 3.Gingerols exhibit inhibitive effect on increase in ileal 5-HT release induced by cisplatin.The antiemetic mechanism of gingerols may refer to the peripheral or central systems of 5-HT and 5-HT3 receptors,DA and DA receptors.
