Objective:Investigate the relationship between bcl-2 gene translocation,amplification and BCL-2 protein expression,and the role of bcl-2 gene abnormal in classification of diffuse large B-cell lymphoma(DLBCL).Method:Two hundred and twenty cases of DLBCL were selected in this investigation.Tissue microarray and in-situ fluorescence hybridization(FISH) technique were used to detect bcl-2 gene translocation and amplification.The phenotypes of either germinal certer B-cell-like(GCB)or non-germinal center B-cell-like(non-GCB)were determined by immunohistochemistry including CD10,BCL-6 and MUM1(EliVISionTM method)using the tissue microarray format.The role of bcl-2 gene translocation,amplification in classification and the relationship among different parameters were analyzed by appropriate statistical methods.Results:Among 220 cases of DLBCL,the difference of bcl-2/IGH translocation,bcl-2 gene amplification between GCB group and non-GCB group was statistical significant,but bcl-2/IGH gene translocation was higher in GCB group(75%>25%) and bcl-2 gene amplification was higher in non-GCB group(74.14%>25.86%).bcl-2 gene amplification can lead to an over-expression of BCL-2 protein and Ki-67 protein.,bcl-2/IGH gene translocation have not this effect. Conclusions:(1)FISH is preferred method in detecting chromosome abnormality.(2)bcl-2 gene amplification is higer express in non-GCB group in this region(74.14%).The patients who have bcl-2 gene amplification were always have high expression of Ki67 and BCL-2.It maybe play an important role in non-GCB group lymphoma.(3)The positive of bcl-2/IGH gene translocation was 3.64%in this region,which is lower than the proportion of the same classification research in abroad,whether it's the peculiar represent of this area's DLBCL need more research.
