Objective : To detect the expression levels of CD4~+CD25~+Treg and FOXP3 gene in aplastic anemia and myelodysplastic syndrome patients, and to study its Clinical significance .Methods : Twenty-three patients with aplastic anemia(AA),Twenty-two patients with myelodysplastic syndrome(MDS) and eight normal controls were studied the expression levels of CD4~+CD25~+Treg was detected by the immunofluorescence and flow cytometry . The expression levels of FOXP3 were measured by the real-time quantitative reverse transcription polymerase chain reaction。Reasults : 1. The expression levels of CD4~+CD25~+Treg and FOXP3 in the aplastic anemia group were obviously lower than those of in normal controlã€(6.00±1.33 %) VS (8.35±1.32 %) or 3.45±1.20 VS 6.45±1.39 (P< 0.01)】2.The expression levels of CD4~+CD25~+Treg and FOXP3 in the myelodysplastic syndrome group were not obviously different with the normal controlã€(8.91±2.58%) VS (8.35±1.32%) or 6.63±2.78 VS 6.52±1.50 (P>0.05)】3. The expression levels of CD4~+CD25~+Treg and FOXP3 were increase gradually in RA,RCMD,RAEB groups ;patients with RA group had obviously lower than those in the RCMD,RAEB groups and normal controls ;patients with RAEB had significantly higher CD4~+CD25~+Treg and FOXP3 than the RA,RCMD groups and normal controls .4.In the myelodysplastic syndrome group , the expression levels of CD4~+CD25~+Treg and FOXP3 were obviously higher than those of aplastic anemia groupã€(8.91±2.58%)VS (6.00±1.33%)or 6.63±2.78 VS 3.47±1.47 (P<0.01)】;In the chronic aplastic anemia group , the expression levels of CD4~+CD25~+Treg and FOXP3 were not obviously different in the RA group.5. The levels of CD4~+CD25~+Treg in aplastic anemia group and myelodysplastic syndrome group were positively correlated with those of FOXP3 ,Pearson respective: 0.735,0.630(P﹤0.01)Conclusion : 1. There are exist the closely correlate between the etiopathogenesis of aplastic anemia with the levels of CD4~+CD25~+Treg and FOXP3 gene . Imunologic abnormality is the main etiological factor to the aplastic anemia2. Existing the immune state heterogeneity in the myelodysplastic syndrome group, Immunologic abnormality to promote the advancement of the myelodysplastic syndrome. But patients in the the myelodysplastic syndrome group have ununiformity in immune state . So identify immunization factor Precisely ffectiveness in the myelodysplastic syndrome ,we must carry out more investigation.
